Synthesis and characterization of a crosslinked membrane based on sulfonated poly(aryl ether sulfone) and sulfonated polyvinyl alcohol applied in direct methanol fuel cells

Vinyl-terminated sulfonated poly (arylene ether sulfone) (VSPAES) and sulfonated polyvinyl alcohol containing vinyl group (VSPVA) have been prepared. CpCo(i) precatalysts for [2+2+2] cycloaddition reactions: synthesis and reactivity

The efficient synthesis and structural characterisation of a series of novel CpCo(i)-olefin-phosphite/phosphoramidite complexes and their evaluation in catalytic cyclotrimerisation reactions are reported. Pulsed electron-electron double-resonance (PELDOR) measurements are presented from the potassium ion channel KcsA both solubilized in detergent and reconstituted in lipids. Site-directed spin-labeling using (1-oxyl-2,2,5,5-tetramethyl-3-pyrrolin-3-yl)methyl methanethiosulfonate was performed with a R64C mutant of the protein. The orientations of the spin-labels in the tetramer were determined by PELDOR experiments performed at two magnetic field strengths (0.3 T/X-band and 1.2 T/Q-band) and variable probe frequency. Quantitative simulation of the PELDOR data supports a strongly restricted nitroxide, oriented at an angle of 65 degrees relative to the central channel axis. In general, poorer quality PELDOR data were obtained from membrane-reconstituted preparations compared to soluble proteins or detergent-solubilized samples. One reason for this is the reduced transverse spin relaxation time T-2 of nitroxides due to crowding of tetramers within the membrane that occurs even at low protein to lipid ratios. This reduced T-2 can be overcome by reconstituting mixtures of unlabeled and labeled proteins, yielding high-quality PELDOR data. Identical PELDOR oscillation frequencies and their dependencies on the probe frequency were observed in the detergent and membrane-reconstituted preparations, indicating that the position and orientation of the spin-labels are the same in both environments.

Oxidation of pyrrole by dehaloperoxidase-hemoglobin: chemoenzymatic synthesis of pyrrolin-2-ones

The use of oxidoreductases as biocatalysts in the syntheses of functionalized, monomeric pyrroles has been a challenge owing to, among a number of factors, undesired polypyrrole formation. Here, we have investigated the ability of dehaloperoxidase (DHP), the coelomic hemoglobin from the terebellid polychaete Amphitrite ornata, to catalyze the H2O2-dependent oxidation of pyrroles as a new class of substrate for this enzyme. Substrate oxidation was observed for all compounds employed (pyrrole, N-methylpyrrole, 2-methylpyrrole, 3-methylpyrrole and 2,5-dimethylpyrrole) under both aerobic and anaerobic conditions. Using pyrrole as a representative substrate, only a single oxidation product, 4-pyrrolin-2-one, was observed, and notably without formation of polypyrrole. Reactivity could be initiated from all three biologically relevant oxidation states for this catalytic globin: ferric, ferrous and oxyferrous. Isotope labeling studies determined that the O-atom incorporated into the 4-pyrrolin-2-one product was derived exclusively from H2O2, indicative of a peroxygenase mechanism. Consistent with this observation, single-and doubl-emixing stopped-flow UV-visible spectroscopic studies supported compound I, but not compounds ES or II, as the catalytically-relevant ferryl intermediate involved in pyrrole oxidation. Electrophilic addition of the ferryl oxygen to pyrrole is proposed as the mechanism of O-atom transfer. The results demonstrate the breadth of chemical reactivity afforded by dehaloperoxidase, and provide further evidence for establishing DHP as a multifunctional globin with practical applications as a biocatalyst.

Elevated CO2 improves glucosinolate metabolism and stimulates anticancer and anti-inflammatory properties of broccoli sprouts

Sprouting process enhances plant bioactive compounds. Broccoli (Brassica oleracea L) sprouts are well known for their high levels of glucosinolates (GLs), amino acids, and antioxidants, which offer outstanding biological activities with positive impacts on plant metabolism. Elevated CO2 (eCO(2), 620 ppm) was applied for 9 days to further improve nutritive and health-promoting values of three cultivars of broccoli sprouts i.e., Southern star, Prominence and Monotop. eCO(2) improved sprouts growth and induced GLs accumulation e.g., glucoraphanin, possibly through amino acids production e.g., high methionine and tryptophan. There were increases in myrosinase activity, which stimulated GLs hydrolysis to yield health-promoting sulforaphane. Interestingly, low levels of ineffective sulforaphane nitrile were detected and positively correlated with reduced epithiospecifier protein after eCO(2) treatment. High glucoraphanin and sulforaphane levels in eCO(2) treated sprouts improved the anticarcinogenic and anti-inflammatory properties of their extracts. In conclusion, eCO(2) treatment enriches broccoli sprouts with health-promoting metabolites and bioactivities.

Effects of graphene surface functionalities towards controlled reinforcement of a lignin based renewable thermoplastic rubber

This report describes methods to enhance mechanical properties of a renewable thermoplastic rubber by incorporating functional graphene platelets into an equal-mass mixture of lignin, a feedstock from plant biomass, and a nitrile-butadiene rubber having 41 mol % acrylonitrile content (NBR41) that form a nanocomposite. Not all lignins yield mechanically strong material when they are combined with NBR41, and thus, limit its use for such rubbery products. Here, we evaluate different graphene oxides (GO) and reduced graphene oxides (rGO) dry-powders with different surface areas, functionality, and wettability in the synthesis of performance-enhanced nanocomposites of soft lignin/NBR41 matrix via a solvent-free, high shear reactive process. High surface area GO (HSGO) platelets with strong hydrophilicity exhibit good dispersion in the multiphase lignin/NBR41 composites with lignin dispersion varying from 200 to 2000 nm, resulting in superior reinforcement over other graphene derivatives. The addition of 1-4 wt % HSGO increased the tensile stress of the lignin/NBR41 thermoplastic rubber matrix by 60-160% (15-24 MPa) and the modulus by 200-700% (60-140 MPa). Scanning electron microscopy and small angle X-ray scattering results show that the well-dispersed HSGO platelets effectively disrupt the lignin-rich aggregates in NBR41 matrix, resulting in both strength and stiffness enhancements in the formed nanocomposites. This report on performance enhancement of lignin-based renewable polymers via the production of nanocomposite would increase potential for `finding the value of lignin’-a grand challenge associated with modern biorefineries.

Bacterial profiles and volatile flavor compounds in commercial Suancai with varying salt concentration from Northeastern China

Suancai, a popular traditional fermented cabbage in northeast China, is commonly prepared by the spontaneous fermentation process at a certain salt concentration. The salt can affect the metabolites by affecting the microorganisms during Suancai fermentation. The bacterial community and volatile flavor compounds in commercial Suancai from different regions of Northeastern China at different salt concentrations were investigated using next-generation sequencing and GC-MS. Firmicutes and Cyanobacteria were the dominant phyla in the commercial Suancai, and Lactobacillus, Pediococcus, and Leuconostoc were the dominant genera. Among them, Lactobacillus and Pediococcus were considered as the biomarkers of the low and high salt Suncai, respectively. Eighty-five volatile flavor compounds were detected, and HS exhibited higher contents of volatile flavor compounds than LS. Based on the results of correlation analysis, Pediococcus were highly correlated with the alcohols and nitriles in Suancai. The contents of alcohols and nitriles significantly increased in the Suancai, fermented by Pediococcus pentosaceus. The co-inoculated fermentation of Lactobacillus plantarum and P. pentosaceus could increase the concentrations of alcohols, esters, aldehydes, hydrocarbons, and nitriles in Suancai. This study provides a perspective for understanding the ecology of Suancai fermentation and facilitating the fermentation with multispecies inoculation fermentation at an appropriate salt concentration.

Nitrile Oxidation at a Ruthenium Complex leading to Intermolecular Imido Group Transfer

The oxidation of an acetonitrile ligand coordinated to ruthenium is explored in deuterated dimethyl sulfoxide by H-1 NMR spectroscopy. When oxidized with an iodosoarene oxygen atom transfer (OAT) reagent, kinetic studies demonstrate that the nitrile ligand does not dissociate before reacting. Instead, OAT to the central nitrile carbon is implicated (nitrile oxidation) and is further supported by the product of the reaction, N-acyldimethylsulfoximine. The N-acyldimethylsulfoximine likely formed by an imido group transfer reaction from ruthenium to the NMR solvent, and the product was synthesized independently to verify its identity in the reaction. This reaction represents the first time that a nitrile oxidation reaction has resulted in intermolecular imido group transfer to a substrate, presumably through a reactive ruthenium(IV)imido intermediate. This suggests that nitrile oxidation is a plausible route into reactive metal-imido intermediates for amination and aziridination reactions.
