Bioprocessing optimization for efficient simultaneous removal of methylene blue and nickel by Gracilaria seaweed biomass

The pollution of water by heavy metal ions and dyes, particularly from industrial effluents, has become a global environmental issue. Therefore, the treatment of wastewater generated from different industrial wastes is essential to restore environmental quality. The efficiency of Gracilaria seaweed biomass as a sustainable biosorbent for simultaneous bioremoval of Ni2+ and methylene blue from aqueous solution was studied. Optimization of the biosorption process parameters was performed using face-centered central composite design (FCCCD). The highest bioremoval percentages of Ni2+ and methylene blue were 97.53% and 94.86%; respectively, obtained under optimum experimental conditions: 6 g/L Gracilaria biomass, initial pH 8, 20 mg/L of methylene blue, 150 mg/L of Ni2+ and 180 min of contact time. Fourier Transform Infrared Spectroscopy (FTIR) spectra demonstrated the presence of methyl, alkynes, amide, phenolic, carbonyl, nitrile and phosphate groups which are important binding sites involved in Ni2+ and methylene blue biosorption process. SEM analysis reveals the appearance of shiny large particles and layers on the biosorbent surface after biosorption that are absent before the biosorption process. In conclusion, it is demonstrated that the Gracilaria seaweed biomass is a promising, biodegradable, ecofriendly, cost-effective and efficient biosorbent for simultaneous bioremoval of Ni2+ and methylene blue from wastewater effluents.

Discovery and Surprises with Cyclizations, Cycloadditions, Fragmentations, and Rearrangements in Complex Settings

We discuss a number of synthesis routes to complex natural products recently reported from our group. Although the structures are quite varied, we demonstrate the research endeavor as a setting to examine the implementation of cyclizations, cycloadditions, rearrangements, and fragmentations. We showcase how the various transformations enabled access to key core structures and thereby allowed the rapid introduction of complexity. Two different routes to (-)-mitrephorone A, the first case discussed, led to the use of Koser’s reagent to effect oxetane formation from diosphenol derivatives. Even though the Diels-Alder cycloaddition reaction represents one of the workhorses of complex molecule synthesis, there are opportunities provided by the complexity of secondary metabolites for discovery, study, and development. In our first approach to (-)-mitrephorone A, Diels-Alder cycloaddition provided access to fused cyclopropanes, while the second synthesis underscored the power of diastereoselective nitrile oxide cycloadditions to access hydroxy ketones. The successful implementation of the second approach required the rigorous stereocontrolled synthesis of tetrasubstituted olefins; this was accomplished by a highly stereoselective Cr-mediated reduction of dienes. The diterpenoid (+)-sarcophytin provided a stage for examining the Diels-Alder cycloaddition of two electron-deficient partners. The study revealed that in the system this unusual combination works optimally with the E,Z-dienoate and proceeds through an exo transition state to provide the desired cycloadduct. Our reported pallambin synthesis showcased the use of fulvene as a versatile building block for the core structure. Fulvene decomposition could be out competed by employing it as a diene and using a highly reactive dienophile, which affords a bicyclic product that can in turn be subjected to chemo- and stereoselective manipulations. The synthesis route proceeds with a C-H insertion providing the core structure en route to pallambin A and B. The studies resulting in our synthesis of gelsemoxonine highlight the use of the acid-catalyzed rearrangement/chelotropic extrusion of oxazaspiro[2.4]heptanes to access complex beta-lactams, which are otherwise not readily prepared by extant methods in common use. Mechanistic investigations of the intriguing ring contraction supported by computational studies indicate that the reaction involves a concerted cleavage of the N-O bond and cyclopropane ring opening under the extrusion of ethylene. The synthesis of guanacastepenes focused on the use of cyclohexyne in [2+2]-cycloadditions with enolates. The resulting cyclobutene can be enticed to undergo ring opening to give a fused six-seven ring system. The cycloinsertion reaction of cyclohexyne developed for the first time proves useful as a general approach to complex fused ring systems.

Influence of the ancillary ligands on the luminescence of platinum(II) complexes with a triazole-based tridentate C boolean OR N boolean OR N luminophore

The effect of different ancillary ligands and counterions in platinum(II) complexes has been investigated. Based on the previously reported tridentate C<^>N<^>N ligand precursor 2-(1-benzyl-1H-1,2,3-triazol-4-yl)-6-phenylpyridine (HL), the photophysical properties of complexes of the type [Pt(L)(X)](n+) have been varied by changing the fourth (monodentate) ligand (X) of the square-planar platinum(II) complexes. Different lifetimes and quantum yields were observed, depending on the identity of this ancillary ligand. The most favorable photophysical properties within this series of complexes were obtained for neutral complexes with the phenylacetylido ligand with a quantum yield of 35% and a lifetime of 2.22 mu s, while for cationic complexes bearing nitrile, isonitrile and triphenylphosphane units gave comparable results with quantum yields ranging from 11% to 16% and lifetimes from 3.59 mu s to 4.93 mu s. Introducing a ferrocene moiety attached to an acetylido ligand, the complex became hardly emissive. The investigated counterions perchlorate, tetrafluoroborate and hexafluorophosphate of positively charged complexes regarding their photophysical properties were found to affect the non-radiative decay rates. To understand the minor effect observed for the emission maxima of the complexes, density functional theory (DFT) was applied. The experimental emission spectra of the complexes were reproduced by using simplified model systems. The distribution of the frontier orbitals used for the description of the emissive T-1 state in its optimized geometry mainly involves the tridentate luminophore rather than the ancillary ligand. This explains why the emission is dominated by the pincer unit with perturbative participation of the metal center while excluding significant influence of the ancillary ligand.

Discovery of 3-hydroxy-3-pyrrolin-2-one-based mPGES-1 inhibitors using a multi-step virtual screening protocol

Targeting microsomal prostaglandin E-2 synthase-1 (mPGES-1) represents an efficient strategy for the development of novel drugs against inflammation and cancer with potentially reduced side effects. With this aim, a virtual screening was performed on a large library of commercially available molecules using the X-ray structure of mPGES-1 co-complexed with a potent inhibitor. Combining fast ligand-based shape alignment, molecular docking experiments, and qualitative analysis of the binding poses, a small set of molecules was selected for the subsequent steps of validation of the biological activity. Compounds 2 and 3, bearing the 3-hydroxy-3-pyrrolin-2-one nucleus, showed mPGES-1-inhibitory activity in the low micromolar range. These data highlighted the applicability of the reported virtual screening protocol for the selection of new mPGES-1 inhibitors as promising anti-inflammatory/anti-cancer drugs.
