ULTRASOUND IN FATTY-ACID CHEMISTRY – SYNTHESIS OF A 1-PYRROLINE FATTY-ACID ESTER ISOMER FROM METHYL RICINOLEATE

A novel 1-pyrroline fatty acid ester isomer [viz. 8-(5-hexyl-1-pyrrolin-2-yl)octanoate] has been synthesized from methyl ricinoleate by two routes with an overall yield of 42 and 30%, respectively. Most of the reactions are carried out under concomitant ultrasonic irradiation (20 KHz, ca. 53 watts/cm(2)). Under such a reaction condition, the reaction time is considerably shortened, and product yields are high. Dehydrobromination under concomitant ultrasonic irradiation of methyl 9,10-dibromo-12-hydroxyoctadecanoate with KOH in EtOH furnishes methyl 12-hydroxy-9-octadecynoate (66%) within 15 min. Hydration of the latter under ultrasound with mercury(II)acetate in aqueous tetrahydrofuran yields exclusively methyl 12-hydroxy-9-oxo-octadecanoate (95%) in 30 min. The hydroxy group in the latter compound is transformed to the azido function via the mesylate, and treatment of the azido-oxo intermediate (methyl 12-azido-9-oxooctadecanoate) with Ph(3)P under ultrasonic irradiation furnishes the requisite 1-pyrroline fatty acid ester (77%). The same azido-oxo intermediate has also been obtained by the oxidation of methyl 12-azido-9-cis-octadecenoate using benzoquinone and a catalytic amount of Pd(II)chloride in aqueous tetrahydrofuran under concomitant ultrasonic irradiation (90 min) to give the product in 45% yield. The latter reaction does not take place even the reaction mixture.

Insights on the Pseudo-Enantiomeric Properties of Bifunctional Cinchona Alkaloid Squaramide-Derived Organocatalyst

The use of pseudo enantiomers is a well-known method of achieving products of complementary stereochemistry. Only rarely can different enantiomers of a product be accessed without modulation of the catalyst. Recently, a system was reported wherein two different enantiomers of spirocycles were obtained by a cascade reaction of unsaturated pyrrolin-4-ones with mercaptoacetaldehyde catalyzed by a single optimized cinchona alkaloid squaramide-derived organocatalyst. It was originally proposed that the E/Z geometry of the unsaturated pyrrolin-4-one dictated the stereochemistry of the spirocycle product, but this was not investigated further. In the present work, we have investigated the nature of a pseudo-enantiomeric organocatalyst conformation applying density functional theory calculations for investigating the transition states for the reaction. Furthermore, the influence of the double-bond geometry of the pyrrolin-4-one has been studied beyond what is possible to test experimentally. The results provide a greater understanding for this class of reactions that may be applicable in future methodology development.

Fluorescent enzyme-coupled activity assay for phenylalanine ammonia-lyases

Phenylalanine ammonia-lyases (PALs) catalyse the non-oxidative deamination of l-phenylalanine to trans-cinnamic acid, while in the presence of high ammonia concentration the reverse reaction occurs. Herein, we describe the development of an enzyme-coupled fluorescent assay applicable for PAL-activity screens at whole cell level, involving decarboxylation of trans-cinnamic acid (the product of the PAL reaction) by ferulic acid decarboxylase (FDC1) and a photochemical reaction of the produced styrene with a diaryltetrazole, that generates a detectable, fluorescent pyrazoline product. The general applicability of the fluorescent assay for PALs of different origin, as well as its versatility for the detection of tyrosine ammonia-lyase (TAL) activity have been also demonstrated. Accordingly, the developed procedure provides a facile tool for the efficient activity screens of large mutant libraries of PALs in presence of non-natural substrates of interest, being essential for the substrate-specificity modifications/tailoring of PALs through directed evolution-based protein engineering.

The Morita-Baylis-Hillman (MBH) reaction and [3, 3]-sigmatropic rearrangement are two paradigms in organic synthesis. We have merged the two types of reactions to achieve [3,3]-rearrangement of aryl sulfoxides with alpha,beta-unsaturated nitriles. The reaction was achieved by sequentially treating both coupling partners with electrophilic activator (Tf2O) and base, offering an effective approach to prepare synthetically versatile alpha-aryl alpha,beta-unsaturated nitriles with Z-selectivity through direct alpha-C-H arylation of unmodified alpha,beta-unsaturated nitriles. The control experiments and DFT calculations support a four-stage reaction sequence, including the assembly of Tf2O activated aryl sulfoxide with alpha,beta-unsaturated nitrile, MBH-like Lewis base addition, [3,3]-rearrangement, and E1cB-elimination. Among these stages, the Lewis base addition is diastereoselective and E1cB-elimination is cis-selective, which could account for the remarkable Z-selectivity of the reaction.

One-Pot Synthesis of B-Aryl Carboranes with Sensitive Functional Groups Using Sequential Cobalt- and Palladium-Catalyzed Reactions

Mapping Io’s Surface Composition With Juno/JIRAM

The surface composition of Io is dominated by SO2 frost, plus other chemical species identified or proposed over the past decades by combining Earth-based and space-based observations with laboratory data. Here we discuss spectroscopic data sets of Io obtained by the Jovian InfraRed Auroral Mapper (JIRAM) spectro-imager onboard Juno in nine orbits, spanning a 3-year period. We display average spectral profiles of Io in the 2-5 mu m range, and we use band depths derived from those profiles to map the geographic distribution of SO2 frost and other spectral features. This data set allows for an similar to 22% surface coverage at 58 to 162 km/px and in a broad range of latitudes. Our results confirm the broadly regional SO2-frost trends already highlighted by Galileo/NIMS. Io’s average spectral profiles as well as the mapping of the 4.47-mu m band also confirm that SO2 exists in the (SOO)-S-32-O-16-O-18 isotopic form. Surprisingly, the mapping performed by JIRAM shows that the absorption band at 2.1 mu m is unrelated to SO2 frost, while we map for the first time the depth of the 2.65-mu m band, highlighting regions enriched in this absorber, possibly H2S. JIRAM data confirm that the 3.92-mu m band, likely due to Cl2SO2, is largely related to the SO2 distribution. The correlation between Cl2SO2 and ClSO2, possibly revealed at 4.62 mu m, is not equally clear. The simultaneous presence of two very weak spectral features at 4.55 and 4.62 mu m suggests that nitrile compounds or tholins may also be present on the surface.
