Development of a Drone-Based Thin-Film Solid-Phase Microextraction Water Sampler to Facilitate On-Site Screening of Environmental Pollutants

Finally, drone TF-SPME sampling of an anthropogenically impacted watercourse indicated that impact from the suspected nearby landfill site was minimal, instead suggesting that internal combustion by-products from vehicles on the nearby Highway 401 played a much larger role in contaminating the watercourse. This conclusion was supported by the confirmed presence of BTEX, styrene, isopropylbenzene, propylbenzene, and 1,3,5-trimethylbenzene. In addition to immediately identifying these compounds on-site using portable gas chromatography-mass spectrometry (GC-MS), samples were taken back to the laboratory for benchtop analysis, further supporting this conclusion.

Quenching of pyrene derivatives’ fluorescence by nitroxide radicals in sodium dodecyl sulfate micellar solutions

The mean aggregation number values are comparable with the literature data only when the partition coefficient of the quencher is higher than 1100 M-1. It is shown that the dynamic fluorescence quenching for the PBA/TEMN pair cannot be described by the Infelta-Tachiya model owing to the fact that the intramicellar quenching rate constant is lower than the exit rate constant of the quencher from the micelle. The average location of the fluorescent probes is also discussed, Py and PDA having the pyrenyl moieties located at approximately the same depth in the micellar core, while in the case of PBA the pyrenyl moiety is buried deeper. Machine learning-based prediction of enzyme substrate scope: Application to bacterial nitrilases

Predicting the range of substrates accepted by an enzyme from its amino acid sequence is challenging. Although sequence- and structure-based annotation approaches are often accurate for predicting broad categories of substrate specificity, they generally cannot predict which specific molecules will be accepted as substrates for a given enzyme, particularly within a class of closely related molecules. Combining targeted experimental activity data with structural modeling, ligand docking, and physicochemical properties of proteins and ligands with various machine learning models provides complementary information that can lead to accurate predictions of substrate scope for related enzymes. Here we describe such an approach that can predict the substrate scope of bacterial nitrilases, which catalyze the hydrolysis of nitrile compounds to the corresponding carboxylic acids and ammonia. Each of the four machine learning models (logistic regression, random forest, gradient-boosted decision trees, and support vector machines) performed similarly (average ROC = 0.9, average accuracy = similar to 82%) for predicting substrate scope for this dataset, although random forest offers some advantages. This approach is intended to be highly modular with respect to physicochemical property calculations and software used for structural modeling and docking.
