Stable free radicals belonging to different classes have been synthesized in view of potential applications in studies of organized systems. Among them a new pyrroline nitroxide with a high fluorine content, 2,2,5,5-tetramethyl-3-pyrrolin-1-yloxy-3-[(1H,1H-perfluoro-1-octoxy)carbonyl] (3), apt to probe the hydrophobic perfluoro-rich domains inside macroaggregates, has been prepared by a rarely encountered approach namely the spin-labelling of the very cosurfactant participating in the self-assembling process. Emphasis has then been laid upon the synthesis of further stable free radicals from the group of substituted nitronyl nitroxides. A general experimental procedure has been adapted according to the nature of the 2-substituent, resulting in a series of nitronyl nitroxides with a wide range of hydrophobicity and showing spectral parameters in good agreement with literature values. Using the spin probe-electron spin resonance technique, 3 and the 1-H-imidazol-1-yloxy-4, 5-dihydro-4,4,5, 5-tetramethyl-2-ethyl-3-oxide (7, R = C2H5) or the 1-H-imidazol-1-yloxy-4,5-dihydro-4,4,5,5 (7, R = C11H23) have been applied in ESR investigations on vesicle phases of a zwitterionic surfactant (TDMAO)/perfluorocosurfactant (PFC) system and on Triton X-100 reverse micelles, respectively. While spin probe 3 proved to be insensitive to differences between the charged and uncharged phases, probes 7 were able to detect changes in the micelle micropolarity induced by hydration. Synthesis and evaluation of the anti-proliferative activity of diaryl-3-pyrrolin-2-ones and fused analogs

Analogs containing a central 3-pyrrolin-2-one core with different methoxyphenyl and/or indole substituents were prepared and tested for anti-proliferative activity in U-937 cells. The most efficacious analogs were non-rigid, (non-fused) contained methoxyaryl groups located at the 4-position, and contained either methoxyaryl or indole groups located at the 3-position. Both the number of methoxy groups contained in the substituents and the particular location of the indole rings with respect to the lactam carbonyl had significant affects on anti-proliferative activity. Gein, a new biologically active compound, a 3-pyrrolin-2-one derivative – KOH-1 was synthesized. This compound is at the preclinical research stage now. The aim of this work was the development of methods for determination of KOH-1 in urine by high performance liquid chromatography (HPLC), the study of excretion KOH-1 from the organism of laboratory animals. Materials and methods. Studies on the development of methods were carried out by using a liquid chromatograph LC-20 Prominence (Shimadzu, Japan) with a diode-array detector. The validation was carried out in accordance with the requirements for bioanalytical methods, in terms of selectivity, linearity, precision and accuracy. The study of excretion of KOH-1 was performed on white non-linear male rats weighing 300-400 g. The substance KOH-1 was administered once orally in a suspension of starch mucus at a dose of 100 mg/kg. Results and discussion. As a result of the research, the method for determining the biologically active compound KOH-1 in urine has been developed. The validation showed its suitability for pharmacokinetic studies. The data on daily excretion of KOH-1 in urine after a single oral administration to rats were obtained. Conclusion. The developed conditions for the chromatographic determination of KOH-1 in urine can be used in pharmacokinetic studies, both at the preclinical and clinical stages of the study of a potential drug. The data on excretion of KOH-1 will allow to determine the ways of excretion of the preparation, and also to select a rational dosage, to identify possible contraindications to the use.

A comparative study on mechanical and rheological properties of ternary rubber blends

Polymer blends lead to producing a new class of plastics, which may have better properties than being individual. In this study, the ternary blends of natural rubber (NR)/styrene-butadiene rubber (SBR)/nitrile rubber (NBR) were prepared via melt compounding, and then the physicomechanical properties of ternary NR/SBR/NBR blends were investigated. NR content was kept constant at 30 phr, while SBR and NBR fractions were varied simultaneously. The effect of the addition of 3 phr of ultrablend 4000 as compatibilizer on the compatibility of the rubber blends was studied. The results revealed that the use of the compatibilizer has significantly resulted in the clear stability of the scorch time and the optimum cure time of the blends. Tensile strength; stress at 100%, 200%, and 300% elongations; elongation at break %; compression strength; and compression set increase with the NBR content. There was a dramatic rise in the value of swelling ratio with the decrease of NBR. Moreover, the dynamic mechanical analysis showed some shifts in glass transition temperatures for blends to higher and lower temperatures as a function of the composition, which indicates the partial miscibility between the contributing components. Scanning electron microscopy of the tear fracture surfaces indicated that incorporation of NBR in the blends resulted in better adhesion of ternary blends and improved the tensile properties of ternary blends.

DFT (Density Functional Theory) calculations on the supramolecular dimers in the crystal structure of 1 reveal that the sum of contributions of anion-pi, pi-pi and other long range interactions due to the approximation of the bulk monomers is energetically significant. Molecular Electrostatic Potential (MEP) surface and Quantum Theory of Atoms in Molecules (QTAIM) analyses on the interesting supramolecular dimers of the crystal structures of 2 reveal the presence of unconventional anion center dot center dot center dot pi contacts involving coordinated chlorido ligands and C-H center dot center dot center dot pi(nitrile) interactions involving the pi-system of the nitrile moiety of 3-cyanopyridine. SYNTHESIS AND PHARMACOLOGICAL ACTIVITY OF 1-ALKOXYARYL-5-ARYL-4-ACYL-3-HYDROXY-3-PYRROLIN-2-ONES

A series of 1-alkoxyaryl-5-aryl-4-acyl-3-hydroxy-3-pyrrolin-2-ones were synthesized by the reaction of methyl esters of acylpyruvic acids with a mixture of aromatic aldehyde and 2-, 3-, and 4-methoxyanilines. Photoactivated Refractive Index Anisotropy in Fluorescent Thiophene Derivatives

The optical control of anisotropy in materials is highly advantageous for many technological applications, including the real-time modulation of another light signal in photonic switches and sensors. Here, we introduce three thiophene derivatives with a donor-acceptor structure, which feature different positions of an electron-acceptor nitrile group, and both photoalignment and luminescence properties. Quantum chemical calculations highlight the presence of trans-forms stable at room temperature and metastable cis-isomers. Besides photoluminescence peaked at 440460 nm and 0.4 ns lifetime, the three nonlinear optical chromophores exhibit photoinduced anisotropy of the refractive index closely depending on the specific molecular structure, with higher values of birefringence at lower driving signal being obtained for ortho substitution of the nitrile group. All-optical modulation of an external light beam at rates of hundreds of hertz is demonstrated in the fluorescent systems. This finding opens an interesting route to multispectral photonic switches embedded in the active layers of light-emitting devices.
