Identification of the C4 ‘-Oxidized Abasic Site as the Most Abundant 2-Deoxyribose Lesion in Radiation-Damaged DNA Using a Novel HPLC-Based Approach

A novel analytical high-performance liquid chromatography (HPLC)-based method of quantification of the yields of C4′-oxidized abasic sites, 1, in oxidatively damaged DNA has been elaborated. This new approach is based on efficient conversion of 1 into N-substituted 5-methylene-Delta(3)-pyrrolin-2-ones, 2, upon treatment of damaged DNA with primary amines in neutral or slightly acidic solutions with subsequent quantification of 2 by HPLC. The absolute and relative radiation-chemical yields of 1 in irradiated DNA solutions were re-evaluated using this method. The yields were compared with those of other 2-deoxyribose degradation products including 5-methylene-2(5H)-furanone, malondialdehyde, and furfural resulting from the C1′, C4′ and C5′-oxidations, respectively. The yield of free base release (FBR) determined in the same systems was employed as an internal measure of the total oxidative damage to the 2-deoxyribose moiety. Application of this technique identifies 1 as the most abundant sugar lesion in double-stranded (ds) DNA irradiated under air in solution (36% FBR). In single-stranded (ss) DNA this product is second by abundance (33% FBR) after 2-deoxyribonolactones (C1′-oxidation; 43% FBR). The production of nucleoside-5′-aldehydes (C5′-oxidation; 14% and 5% FBR in dsDNA and ssDNA, respectively) is in the third place. Taken together with the parallel reaction channel that converts C4′-radicals into malondialdehyde and 3′-phosphoglycolates, our results identify the C4’-oxidation as a prevalent pathway of oxidative damage to the sugar-phosphate backbone (50% or more of all 2-deoxyribose damages) in indirectly damaged DNA. Targeted development of hydrophilic porous polysulfonamide gels with catalytic activity

We report the use of a template and functional monomers in the synthesis of three novel polysulfonamide gels with new architectures and functional groups. These mesoporous polysulfonamide gels were prepared by the condensation polymerization of benzene-1,3-disulfonyl chloride (as the main precursor), linear monomers, and cross-linkers (as variable precursors) in the presence of a silica template by a combination of sol-gel chemistry and the nanocasting technique. In this synthesis pathway, in situ polymerization onto the template surface led to the construction of a silica/polymer nanocomposite. Next, after removal of the template, the nanocomposite gels were transformed into mesoporous polysulfonamide nanospheres. After the physicochemical identification of the synthesized materials, functionalized polysulfonamides were used as reusable novel catalysts with high efficiency for the Strecker reaction under mild conditions. These polymers have Brunsted/Lewis acid active sites, a mesoporous structure, and hydrogen bonding. Moreover, since these polymers are hydmgels that can absorb water, they can promote the Strecker reaction through chemical absorption of the generated water as a driving force. Overall, this article describes a novel synthesis procedure and application of porous polysulfonamide gels.

Oxidation of pyrrole by dehaloperoxidase-hemoglobin: chemoenzymatic synthesis of pyrrolin-2-ones

The use of oxidoreductases as biocatalysts in the syntheses of functionalized, monomeric pyrroles has been a challenge owing to, among a number of factors, undesired polypyrrole formation. Here, we have investigated the ability of dehaloperoxidase (DHP), the coelomic hemoglobin from the terebellid polychaete Amphitrite ornata, to catalyze the H2O2-dependent oxidation of pyrroles as a new class of substrate for this enzyme. Substrate oxidation was observed for all compounds employed (pyrrole, N-methylpyrrole, 2-methylpyrrole, 3-methylpyrrole and 2,5-dimethylpyrrole) under both aerobic and anaerobic conditions. Using pyrrole as a representative substrate, only a single oxidation product, 4-pyrrolin-2-one, was observed, and notably without formation of polypyrrole. Reactivity could be initiated from all three biologically relevant oxidation states for this catalytic globin: ferric, ferrous and oxyferrous. Isotope labeling studies determined that the O-atom incorporated into the 4-pyrrolin-2-one product was derived exclusively from H2O2, indicative of a peroxygenase mechanism. Consistent with this observation, single-and doubl-emixing stopped-flow UV-visible spectroscopic studies supported compound I, but not compounds ES or II, as the catalytically-relevant ferryl intermediate involved in pyrrole oxidation. Electrophilic addition of the ferryl oxygen to pyrrole is proposed as the mechanism of O-atom transfer. The results demonstrate the breadth of chemical reactivity afforded by dehaloperoxidase, and provide further evidence for establishing DHP as a multifunctional globin with practical applications as a biocatalyst.
