An EPR study of wheat seeds by the use of nitroxide spin probes

The viability of a variety of wheat seeds Mas investigated by use of the EPR technique, Three different types of spin probes (4-Hydroxy-TEMPO (TANOL), 3-Carbamoyl-2,2,5,5-tetramethyl-3-pyrrolin-1-yloxy, 3-Carbamoyl-PROXYL) were utilized in examining the viability of embryo cells. The penetration of the spin probe across the membrane was observed via the change in signal intensity against time for dry embryos of the variety of wheat seeds soaked in aqueous solutions of spin probe and line broadening agent. From the analysis of the signal intensity-time curves (i.e. rehydration curves) it was observed that the two kinds having different genetic roots (Bezostaya, Kunduru) exhibit similar saturation behaviour whereas Gerek saturates much more easily. The rehydration curves for naturally aged seeds indicates that aging causes rapid saturation and an important decrease in the signal intensity. In order to get better insight on the effect of the signals from polar and nonpolar regions on the experimental spectra, these spectra were simulated by the use of the theoretical models developed.

Methylenimine and cyanomethanimine synthesis from ion irradiation of N-2-CH4 ice: Implication on the formation of prebiotic molecules in outer solar system bodies

The synthesis of methylenimine and cyanomethanimine from ion irradiation of N-2-CH4 ice was studied, in an attempt to simulate the role of medium mass cosmic rays and energetic solar particles in the processing of nitrogen-rich ices on cold astrophysical environments, such as those in the outer region of the solar system (e.g. Pluto, Charon, Triton, Makemake and Titan). The N-2-CH4 (90:10) ice mixture was irradiated at 9 K by 38.4 MeV Ca-40(9+) (0.96 MeV/u) at the GANIL facility (Caen/Fance). The evolution of the samples was monitored using insitu Fourier transform infrared spectroscopy (FTIR). The results indicate the formation of CH2NH and CH2NCN, which are considered species of interest in prebiotic chemistry. Other species produced by radiolysis were HCN, HNC, hydrocarbons and nitriles. Direct comparison of the laboratory spectrum from the mixture of reaction products provides an efficient way to focus on the identification of chemical synthesis routes for the production of molecules important in the development of life that are consistent with the chemical inventory and physical conditions on frozen moons and cold objects in the outer solar system.

Ethanol amine functionalized electrospun nanofibers membrane for the treatment of dyes polluted wastewater

This study investigated adsorption kinetics, adsorption equilibrium, and adsorption isotherm of three dyes [i.e., methylene blue (MB), rhodamine-B (RB), and safranin T (ST)] onto polyacrylonitrile (PAN) and ethanolamine (EA) grafted PAN nanofibers (NFs) membranes (EA-g-PAN). The membranes were characterized by field emission scanning electron microscopy (FE-SEM), Fourier-transform infrared spectroscopy (FT-IR) spectroscopy, and Brunauer-Emmet-Teller (BET). FE-SEM showed a smooth morphology for the NFs before and after grafting, while FT-IR confirmed EA grafting into the nitrile group of PAN. The grafting percentage with no change in the physical nature of the membrane was 12.18%. The nitrogen adsorption-desorption isotherms for PAN and EA-g-PAN NFs membranes were similar and classified as a Type IV according to the International Union of Pure and Applied Chemistry. The surface area, pore-volume, and pore size of the EA-g-PAN increased to 21.36 m(2) g(-1), 0.16 cm(3) g(-1), and 304.93 angstrom, respectively. The pores were cylindrical mesopores with bimodal openings, which means that pores were open at both ends. The adsorption of the MB, RB, and ST dyes onto the PAN and EA-g-PAN NFs membranes leveled off at similar to 60 min. The adsorption kinetics showed good fitting to pseudo-second-order kinetic model and multi-step diffusion process. The order of the dye adsorption was PAN < EA-g-PAN. The data were fitted to Langmuir and Freundlich models. The correlation coefficient (r(2)) for Langmuir ranged from 0.940 to 0.995, whereas that for Freundlich ranged from 0.941 to 0.998. After adsorption, FTIR only showed hydrogen bonding between the dyes and the membrane. The reusability experiments showed max desorption for MB (94%), RB (92%), and ST (85%). The EA-g-PAN NFs membrane showed potential for the removal of organic pollutants from the environment. MODULATING ACTION OF VITAMIN C ON THE CYTOTOXIC EFFECT OF MYOSMINE

Myosmine (3-(1-pyrrolin-2-yl)pyridine) is an alkaloid found in tobacco as well as in various foods, fruits and vegetables. Myosmine has recently been suspected to be a tobacco-independent carcinogenic source connected to the increase in incidence of esophageal adenocarcinoma. It has been shown that reactions of nitrosation and peroxidation of myosmine take part in its metabolic activation and leads to the formation of the esophageal carcinogens N’-nitrosonornicotine and 4-hydroxy-1-(3-pyridy1)-1-butanone. Increased cancerogenic potential of myosmine may be expected when the oxidative stress in the cell is enhanced. In order to examine the relationship between the oxidative status of the cells and the inhibitory action of myosmine on the cell growth we studied the combined effect of the antioxidant vitamin C and myosmine on cell proliferation as well as on the cell cycle of murine erythroleukemia (MEL) cells. Protective effect of vitamin C was found at incubation time of 48 hours. It was clearly observed that vitamin C had reduced the toxicity of myosmine in a wide range of concentrations. In order to see how cell cycle is affected in the presence of vitamin C and myosmine we conducted flow cytometric analysis. A combination of vitamin C with 100 mu M myosmine resulted in elimination of the effect of myosmine, respectively the histogram was almost identical to that of the control untreated cells. This result showed that the antioxidant had reduced the influence of myosmine on the cell cycle of MEL. At higher myosmine concentration (200 mu M) the addition of vitamin C reduced the amount of hypoploid cells almost in half. This could be connected with the smaller fraction of apoptotic cells. The conclusion can be drawn that vitamin C may protect cells from the cytotoxic effect of myosmine.

Quantum chemical study on the formation of isopropyl cyanide and its linear isomer in the interstellar medium

The formation mechanism of linear and isopropyl cyanide (hereafter n-PrCN and i-PrCN, respectively) in the interstellar medium (ISM) has been proposed from the reaction between some previously detected small cyanides/cyanide radicals and hydrocarbons/hydrocarbon radicals. n-PrCN and i-PrCN are nitriles therefore, they can be precursors of amino acids via Strecker synthesis. The chemistry of i-PrCN is especially important since it is the first and only branched molecule in ISM, hence, it could be a precursor of branched amino acids such as leucine, isoleucine, etc. Therefore, both n-PrCN and i-PrCN have significant astrobiological importance. To study the formation of n-PrCN and i-PrCN in ISM, quantum chemical calculations have been performed using density functional theory at the MP2/6-311++G(2d,p)//M062X/6-311+G(2d,p) level. All the proposed reactions have been studied in the gas phase and the interstellar water ice. It is found that reactions of small cyanide with hydrocarbon radicals result in the formation of either large cyanide radicals or ethyl and vinyl cyanide, both of which are very important prebiotic interstellar species. They subsequently react with the radicals CH2 and CH3 to yield n-PrCN and i-PrCN. The proposed reactions are efficient in the hot cores of SgrB2 (N) (where both n-PrCN and i-PrCN were detected) due to either being barrierless or due to the presence of a permeable entrance barrier. However, the formation of n-PrCN and i-PrCN from the ethyl and vinyl cyanide always has an entrance barrier impermeable in the dark cloud; therefore, our proposed pathways are inefficient in the deep regions of molecular clouds. It is also observed that ethyl and vinyl cyanide serve as direct precursors to n-PrCN and i-PrCN and their abundance in ISM is directly related to the abundance of both isomers of propyl cyanide in ISM. In all the cases, reactions in the ice have smaller barriers compared to their gas-phase counterparts.

Self-assembling in nafion perfluorinated ionomers based on ESR spectra of novel fluorinated nitroxide spin probes

ESR spectra of three fluorinated nitroxide radicals with different lengths of the fluorinated side chain were measured in neat solvents and in aqueous Nafion solutions and membranes swollen by water. The probes were prepared by condensation of the 3-carboxy-2,2,5,5-tetramethyl-3-pyrrolin-1-yloxyl acid chloride (acid probe, AP) with 1H, 1H-perfluoroalkanols, CF3(CF2)(n)CH2OH, where n = 6, 10, and 16. The corresponding notation for the probes is FP8, FP12, and FP18. The N-14 hyperfine splittings (A(zz) and a(N)) are sensitive to the local site: A(zz) in the range 33.3-36.5 G and a(N) in the range 13.95-16.48 G were measured for solvents ranging from perfluorinated n-hexane to 10 M LiCl/water solution. The line shapes in the probe solutions at and near 300 K are sensitive to the presence of oxygen; exceptionally narrow signals (peak-to-peak width 0.1 G) were detected in carefully deaerated probe solutions, thus allowing the measurement of small hyperfine splittings (typically 0.24 G) from the methyl protons. ESR spectra of the fluorinated probes in Nafion solutions and membranes suggested the presence of multiple sites where the probes exhibited a range of dynamics. A possible reason for this effect is the location of probes in a range of amorphous phases where the dynamics is restricted by the proximity to crystalline polymer domains. The A(zz) values for the slow component of the probes in Nafion solutions and in membranes swollen by water indicated the location of the nitroxides in polar sites, where the local polarity is similar to that in the 10 M LiCl/water system. Probes with longer fluorinated segments penetrate deeper into the assembled polymer chains, farther away from the interface between the polymer aggregate and the solvent. Structural information that can be deduced from protiated and fluorinated probes intercalated in Nafion systems was compared, based on present results and previous studies.

Targeted cytotoxic somatostatin analogue AN-238 inhibits somatostatin receptor-positive experimental colon cancers independently of their p53 status

The resistance of advanced colorectal cancers to therapy is often related to mutations in the p53 tumor suppressor gene. Because somatostatin (SRIF) receptors (ssts) are present in colorectal carcinomas, the treatment with targeted cytotoxic SRIF analogue AN-238, consisting of 2-pyrrolin-odoxorubicin (AN-201) linked to octapeptide SRIF carrier RC-121, may overcome this resistance by producing a higher concentration of the cytotoxic agent in the tumors. Four colon cancer cell lines, HCT-116 and LoVo expressing wild-type p53, and HCT-15 and HT-29 with mutated p53, were investigated. HCT-116, HCT-15, and HT-29, but not LoVo possess functional ssts. We analyzed changes in p53, p21, and proliferating cell nuclear antigen (PCNA) concentrations in these cells in vitro by immunoblotting after exposure to AN-238, its radical AN-201, or doxorubicin (DOX). Equitoxic doses of AN-238, AN-201, or DOX affected p53, p21, and PCNA differently. Analysis of the p21:p53 ratios revealed that DOX increased p53 levels, but most of p53 was mutated and inactive, whereas AN-238 produced smaller changes in p53 concentrations but enhanced its activity. In HCT-15 cells, PCNA:p21 ratios, which are indicators of proliferation and repair processes, remained unchanged after exposure to AN-238 but were increased by DOX. In vivo studies in nude mice demonstrated that AN-238, AN-201, and DOX were equally effective on HCT-1 16 tumors that express wild-type p53. However, AN-238 also inhibited the growth of HCT-15 and HT-29 cancers that express mutant p53, whereas AN-201 and DOX showed no effect. None of the compounds could suppress the proliferation of LoVo tumors that lack functional ssts.
