The study of genotoxicity of two newly synthesized pyrrolinone derivatives on L5178Y mouse lymphoma and bone marrow cells

Genotoxic activity of two newly synthesized pyrrolinone derivatives (lactone (PTB) and lactame (PTP)) was studied both in vitro and in vivo. The substances were incubated with L5178Y mouse lymphoma cells for 4 h, then washed from the substances, and incubated for 18 h. PTP induced significantly increased number of micronuclei (MN) only at the highest dose used (1000 mug/ml). The 20-fold increase of the concentration of PTP led to 2.4-fold increase of the number of MN. PTP wasn’t toxic for lymphoma cells in the applied concentration range. In contrast, PTB induced significantly increased number of MN in lymphoma cells beginning from the concentration of 50 mug/ml and was toxic for lymphoma cells at concentrations of 500 mug/ml and higher. In the comet assay with L5178Y mouse lymphoma cells both substances were active at all concentrations used (PTP at concentrations of 50 mug/ml and 100 mug/ml, and PTB at concentrations of 100 mug/ml and 250 mug/ml). Acute toxicity and MN-inducing activity of compounds were studied on Swiss albino mice. PTB was substantially more toxic for mice than PTP (LD50 was 200 mg/kg and 370 mg/kg respectively). In mouse bone marrow polychromatic erythrocytes PTP induced significantly increased number of MN only at a dose equal to 1/2 of LD50. In contrast, PTB was mutagenic at all doses used -1/2, 1/5, and 1/10 of LD50. At equitoxic doses PTB induced more than 2-fold and 3.5-fold increased levels of MN compared with PTP (1/2 and 1/5 of LD50 respectively). The replacement of =NH group to =O group in chemical structure of pyrrolinone derivatives leads to substantial increase in acute toxicity for mice (1.85-fold) and MN induction activity both in vivo and in vitro.

Regulation of p42/p44 MAPK and p38 MAPK by the adenosine A(1) receptor in DDT1MF-2 cells

The mitogen-activated protein kinase (MAPK) family consists of the p42/p44 MAPKs and the stress-activated protein kinases, c-Jun N-terminal kinase (JNK) and p38 MAPK. We have previously reported that the human adenosine A(1) receptor stimulates p42/p44 MAPK in transfected Chinese hamster ovary cells. In this study, we have investigated whether the endogenous adenosine A(1) receptor in the smooth muscle cell line, DDT1MF-2 activates p42/p44 MAPK, JNK and p38 MAPK. The adenosine A(1) receptor agonist N-6-cyclopentyladenosine stimulated time and concentration-dependent increases in p42/p44 MAPK and p38 MAPK phosphorylation in DDT1MF-2 cells. No increases in JNK phosphorylation were observed following adenosine A(1) receptor activation. N-6-cyclopentyladenosine-mediated increases in p42/p44 MAPK and p38 MAPK phosphorylation were blocked by the selective adenosine A(1) receptor antagonist 1,3-dipropylcyclopentylxanthine and following pretreatment of cells with pertussis toxin. Furthermore, adenosine A(1) receptor-mediated increases in p42/p44 MAPK were sensitive to the MAPK kinase 1 inhibitor PD 98059 (2′-amino-3′-methoxyflavone), whereas p38 MAPK responses were blocked by the p38 MAPK inhibitor SE 203580 (4-(4-fluorophenyl)-2-(4-methylsulfinylphenyl)-5-(4-pyridyl)1H-imidazole). The broad range protein tyrosine kinase inhibitors genistein and tyrphostin A47 (alpha -cyano-(3,4-dihydroxy)thiocinnamide) did not block adenosine A(1) receptor stimulation of p42/p44 MAPK. For comparison, insulin-mediated increases in p42/p44 MAPK were blocked by genistein and tyrphostin A47. The Src tyrosine kinase inhibitor PP2 (4-amino-5-(4-chlorophenyl)-7-(t-butyl)pyrazolo[3,4-d]pyrimidine) and the epidermal growth factor receptor tyrosine kinase inhibitor AG1478 (4-(3-chloroanilino)-6,7-dimethoxyquinazoline) also had no effect on adenosine A(1) receptor stimulation of p42/p44 MAPK. In contrast, wortmannin and LY 294002 (2-(4-morpholinyl)-8-phenyl-4H-1-benzopyran-4-one), inhibitors of phosphatidylinositol 3-kinase, attenuated adenosine A(1) receptor stimulation of p42/p44 MAPK phosphorylation. In conclusion, the adenosine A(1) receptor stimulates p42/p44 MAPK through a pathway which appears to be independent of tyrosine kinase activation but involves phosphatidylinositol 3-kinase. Characterization of Dielectric Relaxation Process by Impedance Spectroscopy for Polymers: Nitrile Butadiene Rubber and Ethylene Propylene Diene Monomer

We invented a dispersion analysis program that analyzes the relaxation processes from dielectric permittivity based on a combination of the Havriliak-Negami and conductivity contribution functions. By applying the created program to polymers such as nitrile butadiene rubber (NBR) and ethylene propylene diene monomer (EPDM), several relaxation processes were characterized: an alpha process due to segmental motions of the C-C bond, an alpha ‘ process attributed to fluctuations in the end-to-end dipole vector of the polymer chain, the conduction contribution by the filler observed above room temperature, and secondary relaxation processes beta and gamma of motion for the side group in NBR. In the EPDM specimen, the beta process associated with the rotational motion of the side groups, the alpha process associated with the relaxation of local segmental motion, and the alpha beta process associated with the origin of the beta process at high temperatures above 305 K were observed. The Maxwell-Wagner-Sillars effect and conduction contribution were also presented. The molecular chains responsible for the relaxation processes were assigned by building molecular models of the two polymers. The temperature dependence of the relaxation strength and the shape parameters that characterize the process were investigated. From the temperature-dependent relaxation analysis, the merged alpha beta process, activation energy, and glass transition temperature were determined and compared.

Condensation of (5) with aromatic aldehydes in the presence of glacial acetic acid yielded new Schiff bases (7). N-phthaloylglycyl chloride reacts with (1) in dioxane in the presence of triethylamine to give N-(N’-phthaloyraminoacyl)-4-aminodiphenylselenide (8), hydrazinolyses of which affords N-(aminoacyl)-4-amino-4′-nitrodiphenylselenide (9). Compound (9) undergoes facile condensation with aromatic aldehydes in the presence of piperidine to give Schiff bases (10). EPR spectroscopy detection of active oxygen and free radicals in thylakoids exposed to photoinhibition

High intensity illumination of thylakoids results in the well-characterized impairment of Photosystem Il electron transport (photoinhibition), followed by the degradation of the D1 reaction centre protein. The time course and features of photodamage are different in fully functional thylakoid membranes, when photoinhibition is invoked by impairment of Photosystem II acceptor side electron transport, and in thylakoids which are unable to oxidize water, when the damage is a consequence of inactivation of Photosystem II donor side. In the present study we followed the production of singlet oxygen and free radicals during both types of photoinhibition by EPR spectroscopy. Singlet oxygen was detected by following the formation of 2,2,6,6-tetramethylpiperidine-1-oxyl, a stable nitroxide radical yielded in the reaction of singlet oxygen with the sterically hindered amine 2,2,6,6-tetramethylpiperidine. Free radicals were detected as spin adducts of the spin trap 5,5-dimethyl-1-pyrrolin-N-oxide, and identified on the basis of hyperfine splitting constants of the EPR spectre. We found that (i) singlet oxygen, a non-radical form of active oxygen was detectable only in samples undergoing acceptor side induced photodamage. (ii) The acceptor side induced process was accompanied by the oxygen dependent production of carbon centred (alkyl or hydroxyalkyl) radicals, probably from the reaction of singlet oxygen with histidine residues. (iii) Donor side induced photoinhibition was dominated by hydroxyl radicals, which were produced in anaerobic samples, too. The production rate of these radicals, as well as D1 protein degradation, was dependent on the possibility of electron donation From manganese ions to Photosystem II. The marked distinction between the active oxygen forms produced in acceptor and donor side induced photoinhibition are in agreement with earlier reports on the different mechanism of these processes.
