A Nanomechanical Study on Deciphering the Stickiness of SARS-CoV-2 on Inanimate Surfaces

The SARS-CoV-2 virus that causes the COVID-19 epidemic can be transmitted via respiratory droplet-contaminated surfaces or fomites, which urgently requires a fundamental understanding of intermolecular interactions of the coronavirus with various surfaces. The corona-like component of the outer surface of the SARS-CoV-2 virion, named spike protein, is a key target for the adsorption and persistence of SARS-CoV-2 on various surfaces. However, a lack of knowledge in intermolecular interactions between spike protein and different substrate surfaces has resulted in ineffective preventive measures and inaccurate information. Herein, we quantified the surface interaction and adhesion energy of SARS-CoV-2 spike protein with a series of inanimate surfaces via atomic force microscopy under a simulated respiratory droplet environment. Among four target surfaces, polystyrene was found to exhibit the strongest adhesion, followed by stainless steel (SS), gold, and glass. The environmental factors (e.g., pH and temperature) played a role in mediating the spike protein binding. According to systematic quantification on a series of inanimate surfaces, the adhesion energy of spike protein was found to be (i) 0-1 mJ/m(2) for hydrophilic inorganics (e.g., silica and glass) due to the lack of hydrogen bonding, (ii) 2-9 mJ/m(2) for metals (e.g., alumina, SS, and copper) due to the variation of their binding capacity, and (iii) 6-11 mJ/m(2) for hydrophobic polymers (e.g., medical masks, safety glass, and nitrile gloves) due to stronger hydrophobic interactions. The quantitative analysis of the nanomechanics of spike proteins will enable a protein-surface model database for SARS-CoV-2 to help generate effective preventive strategies to tackle the epidemic.

Regioselective synthesis of pyrrolin-3-ones and 2,3,4,5-tetrahydro[1,3]-oxazines from N-vinylic amidines

Achiral and optically active N-vinylic amidines are obtained by simple addition of amidines to acetylenic esters. Thermal intramolecular cyclization of these Substrates containing a carboxylate group in position 3 gives pyrrolin-3-ones. The enaminone character of these compounds towards propargyl bromide, diethyl azodicarboxylate, diethyl acetylenedicarboxylate, ethyl propiolate and phenyl isocyanate is studied and functionalized pyrrolin-3-one derivatives are obtained. Application of analytical pyrolysis to gain insights into proteins of condensed corn distillers solubles from selective milling technology

Present study evaluates the analytical pyrolysis to unravel the pyrolysate compounds (PyCs), protein & glycosylation indicators in a protein fraction (PF) from condensed corn distillers solubles (CCDS) of novel selective milling technology. Besides, quadratic and linear corre-lations using protein content (%) by Dumas method, toluene signature PyC, and elemental nitrogen (%) were developed. Pyrolysis was performed at temperature of 400 degrees C, where most of the PyCs released from proteins in a lignin-free PF instead of other constituents. The p cresol and methyl 9,12-octadecadienoate were found with the highest area% of 3.2 and 19.3 in PF and CCDS, respectively. Protein derived PyCs such as nitrogen heterocyclics, amines & amides and nitriles were examined 63.93%, 61.90% and 70% greater in PF than CCDS. Moreover, the indicators of protein were toluene, p-cresol, phenol, phenylacetonitrile, 4vinylphenol, whereas that of glycosylated protein were methyl mercaptan, furan derivatives, and hexahydro-3-(phenylmethyl) pyrrolo [1, 2 a] pyrazine-1,4-dione. The absence of diketopiperazines provided evidence of crosslinking and aggregation of protein. The values of the protein content obtained from the quadratic model and Dumas method were found to be statistically non-significant (p < 0.05) imply the model validity. An efficient Cu/functionalized graphene oxide catalyst for synthesis of 5-substituted 1H-tetrazoles

The copper nanoparticles (Cu NPs) and amide functionalized graphene oxide (Cu-Amd-RGO) catalyst were prepared. This prepared catalyst (Cu-Amd-RGO) used for the synthesis of tetrazole derivatives. The catalyst (Cu-Amd-RGO) was characterized by field emission scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FTIR), and X-ray diffraction analysis (XRD). The average particle size of Cu was found to be 7.6 nm. The Cu-Amd-RGO catalyst exhibited excellent catalytic activity and recyclability for synthesis of tetrazoles.

Synthesis and structure determinations of complexes containing a five-membered lactam structure based on organohydrazido(2-) ligands

Acid-catalyzed reactions of the hydrazido(2-) complexes cis,mer-[WX(2)(NNH2)(PMe(2)Ph)(3)] (X=Cl, Br) with phthalaldehyde gave the (phthalimidin-2-yl)imido complexes cis,mer-[WX(2)(NNCH2C6H4CO)(PMe(2)Ph)(3)], via the condensation of the terminal NH2 group with one of the formyl groups and the following cyclization to form a phthalimidine ring. Crystal structure of the chloro complex 3a was unambiguously determined by X-ray analysis. A similar condensation of the hydrazido(2-) complex trans-[WF(NNH2)(dppe)(2)](+) (8a(+); dppe=Ph(2)PCH(2)CH(2)PPh(2)) with phthalaldehyde resulted in the formation of the diazoalkane complex trans-[WF(NN=CHC6H4CHO)(dppe)(2)](+). A loop resonator for slice-selective in vivo EPR imaging in rats

A loop resonator was developed for 300 MHz continuous-wave electron paramagnetic resonance (CW-EPR) spectroscopy and imaging in live rats. A single-turn loop (55 mm in diameter) was used to provide sufficient space for the rat body. Efficiency for generating a radiofrequency magnetic field of 38 mu T/W-1/2 was achieved at the center of the loop. For the resonator itself, an unloaded quality factor of 430 was obtained. When a 350 g rat was placed in the resonator at the level of the lower abdomen, the quality factor decreased to 18. The sensitive volume in the loop was visualized with a bottle filled with an aqueous solution of the nitroxide spin probe 3-carbamoyl-2,2,5,5-tetramethyl-3-pyrrolin-1-yloxy (3-CP). The resonator was shown to enable EPR imaging in live rats. Theoretical investigation of protonated thiophene and two of its nitrile substituted derivatives (2-cyanothiophene and 3-cyanothiophene)

Theoretical and experimental spectroscopic data for protonated cyano-thiophenes (R-CNH+ with R = C4H3S), which are needed for their interstellar search and/or detection, are still lacking in the literature. Considering the high abundance and reactivity of H-3(+) in the interstellar medium (ISM), a quantum chemical investigation on protonated thiophene and two of its nitrile-substituted derivatives (2-cyanothiophene and 3-cyanothiophene) is undertaken for their characterization. The geometrical structures for the title species are calculated at the M06-2X/6-31G(d,p) level of theory, followed by an empirical correction for systematic errors. At the same level of theory, IR and Raman spectra are explored and the rotational parameters are calculated. The proton affinity (PA) of R-CN and the enthalpy, entropy and Gibbs free energy changes (Delta H-r, Delta S-r and Delta(r)G) of the reactions producing R-CNH+ are computed at the G2(MP2) and G3B3 levels of theory and at different temperatures. The PA calculations show that the protonation favors the nitrogen atom, while Delta H-r, Delta S-r, and Delta(r)G reveal the spontaneous reactions producing R-CNH+ and their neutral forms. In addition, quadrupole hyperfine structures are predicted, while the region where the brightest lines fall at different temperatures is discussed. These results are expected to assist astrophysicists and astrochemists in the search for new species in the ISM.
