A sensitive and selective liquid chromatography-tandem mass spectrometry method for simultaneous determination of five isoquinoline alkaloids from Chelidonium majus L. in rat plasma and its application to a pharmacokinetic study

Chelidonium majus L. is one of the most important medicinal plants of the family Papaveraceae. Its pharmacological effects have been primarily attributed to the presence of a number of alkaloids. In the present study, a sensitive and selective liquid chromatography-tandem mass spectrometry method for simultaneous determination of five isoquinoline alkaloids from Chelidonium majus L. was developed and validated. The analytes (protopine, chelidonine, coptisine, sanguinarine and chelerythrine), together with the internal standard (palmatine), were extracted from acidified rat plasma with ethyl acetate-dichloromethane (4:1, v/v). Chromatographic separation was carried out on a Diamonsil C18 column with an isocratic mobile phase consisting of acetonitrile and water (adjusted to pH 2.3 with formic acid) (30:70, v/v) at a flow rate of 0.4 ml/min. Mass spectrometric detection was performed by selected reaction monitoring mode via electrospray ionization source operating in positive ionization mode. The assay exhibited good linearity (r ? 0.9933) for all the analytes. The lower limits of quantification were 0.197-1.27 ng/ml using only 50 mul of plasma sample. The intra- and inter-day precisions were less than 11.9%, and the accuracy was between -6.3% and 9.3%. The method was successfully applied to the pharmacokinetic study of the five alkaloids in rats after intragastric administration of Chelidonium majus L. extract. NIR induced self-healing electrical conductivity polyurethane/graphene nanocomposites based on Diels?Alder reaction

In this work, a crosslinker terminal maleimide groups (EDM) has been synthesized and was used in reactions with the pendant furan groups of polyurethane (PUE) to prepare the self-healing electrically conductive composites based on Diels-Alder reaction, and Ag nanowires (AgNWs) as the conductive network inlaid in the surface layer of the polymer matrix layer. Inspired by the graphene-based materials for its versatility in both material and chemical properties, and we present a new synthetic strategy for the preparation of functional graphene nanosheets with DA group that can be employed in the self-healing polyurethane system. To our best knowledge, it hasn’t been reported yet the structure graphene oxide materials (rmGO) in any systems. The rmGO served as fillers to reinforce the composites and would participated in the DA reactions. The excellent NIR light-induced self-healing property of prepared composites was realized through the photo-thermal effect of rmGO. The composite is capable of both structural and electrical healing via NIR irradiation. The injured samples can be locally repaired with high precision and efficiency without an obvious influence on those uninjured parts. The underlying polymer matrix can reform via the dynamic dissociation/recombination of Diels-Alder bonds to bring the separated areas of the AgNWs layer into contact to restore the conductivity. The NIR light-induced self-healing electrically conductive composites have wide potential for many fields, e.g., soft robots, flexible devices, and health-monitoring.

Reduced graphene oxide grafted by the polymer of polybromopyrroles for nanocomposites with superior performance for supercapacitors

An integrated structure has been designed by grafting the polymer of polybromopyrroles (PPBP) onto reduced graphene oxide (RGO) to produce RGO/PPBP nanocomposites with superior electrochemical performance for supercapacitors. The RGO/PPBP nanocomposites are featured with a high nitrogen content (>9 at%), enhanced degree of graphitization, improved specific surface area, abundant micropores, and a tunable hierarchical structure on the basis of sample characterization by XRD, Raman, FT-IR, XPS, SEM, high-resolution TEM, BET, and scanning probe microscopy (SPM) techniques. The grafting of PPBP onto RGO not only suppresses agglomeration and restacking of RGO but also tailors the growth of PPBP on RGO, producing a developed hierarchical structure beneficial for mass/charge transfer. The synergistic effect between RGO and PPBP ensures superior electrochemical performance of RGO/PPBP. In a three-electrode mode, the typical RGO/PPBP electrode presents a galvanostatic capacitance (Cg) of 256 F g-1 at a current density of 10 A g-1, with a capacitance retention of 99.2% after 10000 cycles in 1 mol L-1 H2SO4. More significantly, the typical RGO/PPBP&3Verbar;RGO/PPBP supercapacitor cell exhibits a high Ccell value of 68 F g-1 at 5 A g-1, with a capacitance retention of 91.9% after 10000 cycles. Also, relatively high energy density values of 13.6, 9.4, and 6.7 W h kg-1 with the corresponding power density of 0.5, 2.5, and 10 kW kg-1 are achieved, enabling the tested cell to stay at the high level for carbon-based supercapacitors with an aqueous electrolyte.

Synergistic effects of Ginseng C. A. Mey and Astragalus membranaceus (Fisch.) Bunge on activating mice splenic lymphocytes detected by microcalorimetry and the underlying mechanisms predicted by in silico network analysis

The aim of the current study was to investigate the combination effect of Ginseng C. A. Mey (ginseng) and Astragalus membranaceus (Fisch.) Bunge (astragalus) which are commonly used for strengthening immune system. In this study, the effects of ginseng, astragalus and their combination on activating mice splenic lymphocytes were studied by microcalorimetry in vitro. The parameters of Pmax, Q and Tmax of heat-production curve in spleen lymphocytes were recorded and analyzed, as well as their association with drug concentration. Through the combination index analysis, it was found that ginseng and astragalus could induce the heat generation of splenic lymphocytes. Network analysis was used to investigate the mechanisms, which showed that their combination may cause immune activation in the body through several signaling pathways such as T cell receptor signaling pathway and PI3?K-Akt signaling pathway.
