A comparative study on mechanical and rheological properties of ternary rubber blends

Polymer blends lead to producing a new class of plastics, which may have better properties than being individual. In this study, the ternary blends of natural rubber (NR)/styrene-butadiene rubber (SBR)/nitrile rubber (NBR) were prepared via melt compounding, and then the physicomechanical properties of ternary NR/SBR/NBR blends were investigated. NR content was kept constant at 30 phr, while SBR and NBR fractions were varied simultaneously. The effect of the addition of 3 phr of ultrablend 4000 as compatibilizer on the compatibility of the rubber blends was studied. The results revealed that the use of the compatibilizer has significantly resulted in the clear stability of the scorch time and the optimum cure time of the blends. Tensile strength; stress at 100%, 200%, and 300% elongations; elongation at break %; compression strength; and compression set increase with the NBR content. There was a dramatic rise in the value of swelling ratio with the decrease of NBR. Moreover, the dynamic mechanical analysis showed some shifts in glass transition temperatures for blends to higher and lower temperatures as a function of the composition, which indicates the partial miscibility between the contributing components. Scanning electron microscopy of the tear fracture surfaces indicated that incorporation of NBR in the blends resulted in better adhesion of ternary blends and improved the tensile properties of ternary blends.

Metabolism of furans in vitro: Ipomeanine and 4-ipomeanol

Ipomeanine (IPN), 4-ipomeanol (4-IPO), 1-ipomeanol (1-IPO), and 1,4-ipomeadiol ( DIOL) are toxic 3-substituted furans found in mold-damaged sweet potatoes. IPN and 4-IPO are the most toxic, but all produce pulmonary toxicity in cattle and rodents, and 4-IPO induces hepatotoxicity in humans. These furans require metabolic activation to elicit toxicity, but the limited information obtained from previous metabolism studies prompted us to initiate the investigation reported here. Our initial studies of 4-IPO metabolism by rat liver microsomes demonstrated that the oxidation of 4-IPO to IPN and reduction to DIOL occurred and that more IPN was metabolized to a reactive species than 4-IPO or DIOL. Incubation of IPN and Gly produced a 2 ‘-pyrrolin-5 ‘-one adduct establishing that IPN was metabolized to an enedial. N-Acetylcysteine reacted with the 5 ‘-aldehyde of the enedial to give two 2 ‘,5 ‘-dihydro-2 ‘-hydroxyfurans stabilized by H bonding between the 2 ‘-OH and 3 ‘-keto group. Reaction of the enedial metabolite of IPN with one GSH gave several adducts including a pyrrole derived from the 1,2-addition of GSH to the 5 ‘-aldehyde as well as two tricyclic 2 ‘-pyrrolines derived from the 1,4-addition of GSH at the 4 ‘-position. The identities of the pyrrole and 2 ‘-pyrroline GSH adducts were confirmed by observation of structurally similar adducts from Cys conjugation with the enedial metabolite of IPN. Several minor adducts from the conjugation of the enedial metabolite of IPN with two GSH were also detected. Mono-GSH and bis-GSH adducts were derived from both the 1,2- and 1,4-addition of GSH to the enedial metabolite of 4-IPO in rat liver microsomal incubations of 4-IPO and GSH. Sequential oxidation of 4-IPO to IPN and then to the enedial metabolite followed by GSH conjugation also occurred in the 4-IPO incubations. The complex structures of the reaction products of the enedial with biological nucleophiles may explain why the many attempts to identify 4-IPO adducts to protein have not been successful.

Conformational Analysis, Spectroscopy and Photochemistry of Matrix-Isolated 1H- and 2H- Tautomeric Forms of Ethyl Tetrazole-5-Carboxylate

The preferred conformations, infrared spectra and photochemistry of matrix-isolated 1H and 2H tautomers of ethyl tetrazole-5-carboxylate (ET5C) were investigated. Experimentally, isolated monomeric forms of the two tautomers were trapped in cryogenic argon matrices (15 K), and studied by infrared spectroscopy. Identification of the conformational species trapped in the matrices was undertaken by combining the experimental spectro-scopic data with results of quantum chemical calculations performed at the DFT(B3LYP)/6-311++G(d,p) level of theory. For each tautomer, two conformers were observed in the matrices, all forms exhibiting the ester group in the anti configuration (C-O-C-C dihedral equal to 180 degrees). In turn, the conformers having a gauche ester group that are predicted by the calculations to be significantly populated in the gas phase equilibrium (room temperature), were found to convert into the anti forms during matrix deposition (conformational cooling). Very interestingly, and contrarily to what is in general found for tetrazoles, the 1H tautomer is the most stable tautomer of ET5C, and strongly predominates in the studied matrices. Narrowband UV irradiation (lambda = 250 nm) was found to lead to tetrazole fragmentation, with release of molecular nitrogen and production of four isomeric photoproducts (diazirine, cyanamide, carbodiimide and nitrile imine), all of them being successfully identified through comparison of the experimental spectroscopic spectra of the irradiated matrices with the DFT(B3LYP)/6-311++G(d, p) predicted infrared spectra of these species. This study also stresses the structural relevance of the N-H//O = C intramolecular interaction between the ring hydrogen atom and the carbonyl group of the substituent in the 1H tautomer of ET5C.

A SPIN-LABEL THAT BINDS TO MYOSIN HEADS IN MUSCLE-FIBERS WITH ITS PRINCIPAL AXIS PARALLEL TO THE FIBER AXIS

We have used an indane-dione spin label (2-[-oxyl-2,2,5,5-tetramethyl-3-pyrrolin-3-yl)methenyl]indane-1,3-dione), designated InVSL, to study the orientation of myosin heads in bundles of chemically skinned rabbit psoas muscle fibers, with electron paramagnetic resonance (EPR) spectroscopy. After reversible preblocking with 5,5′-dithiobis(2-nitro-benzoic acid) (DTNB), we were able to attach most of the spin label covalently and rigidly to either Cys 707 (SH1) or Cys 697 (SH2) on myosin heads. EPR spectra of labeled fibers contained substantial contributions from both oriented and disordered populations of spin labels. Similar spectra were obtained from fibers decorated with InVSL-labeled myosin heads (subfragment 1), indicating that virtually all the spin labels in labeled fibers are on the myosin head. We specifically labeled SH2 with InVSL after reversible preblocking of the SH1 sites with 1-fluoro-2,4-dinitrobenzene (FDNB), resulting in a spectrum that indicated only disordered spin labels. Therefore, the oriented and disordered populations correspond to labels on SH1 and SH2, respectively. The spectrum of SH2-bound labels was subtracted to produce a spectrum corresponding to SH1-bound labels, which was used for further analysis. For this corrected spectrum, the angle between the fiber axis and the principal axis of the spin label was fitted well by a Gaussian distribution centered at theta(0) = 11 +/- 1 degrees, with a full width at half-maximum of Delta theta = 15 +/- 2 degrees. The unique orientation of InVSL, with its principal axis almost parallel to the fiber axis, makes it complementary to spin labels previously studied in this system. This label can provide unambiguous information about axial rotations of myosin heads, since any axial rotation of the head must be reflected in the same axial rotation of the principal axis of the probe, thus changing the hyperfine splitting. Therefore, InVSL-labeled fibers have ideal properties needed for further exploration of myosin head orientation and rotational motion in muscle.
