Design and research of non-contact triboelectric nanogenerator based on changing electrostatic field

Triboelectric nanogenerator (TENG) and its self-powered sensor based on the principles of contact electricity generation and electrostatic induction have important application prospects in the fields of new energy and internet of things (IoT). In the contact separation process of polymer materials with different electronegativity values, due to the transfer of electrons, a changing electrostatic field will be generated in the space around the polymer. In the existing TENG research, the field strength perpendicular to the plane of the friction layer and the electrode layer is mainly used to generate electrostatic induction, and the electric field effect around the polymer is ignored. According to the principle of electrostatic induction, the internal charge of the conductor in the electric field will be redistributed, which provides a way for the conductor to generate an induced electrical signal on the surface of the conductor without contacting the friction material. In this paper, we design a non-contact triboelectric nanogenerator (NC-TENG) based on changing electrostatic field. The influence of the distance between the conductor and the friction material, the induction area of the conductor and the position of the conductor relative to the friction material on the induced electrical output performance are studied when silicone rubber and nitrile rubber are used as a friction material. The results show that the NC-TENG can produce a stable electrical signal output when the conductor is completely separated from the friction material. The induced voltage of NC-TENG decreases with the increase of the distance between the conductor and the friction material, and gradually increases with the increase of the conductor’s induction area. For the friction material with a size of 30 mm x 30 mm, the electrical output of NC-TENG tends to be stable when its conductor area is 60 mm x 45 mm. In addition, the different orientation of the conductor relative to the friction material also has a significant effect on the induced electrical output. The NC-TENG designed in this paper provides a novel electrical output generation mode, which provides a higher possibility for the subsequent research on TENG and the application of self-powered sensors.

Machine learning-based prediction of enzyme substrate scope: Application to bacterial nitrilases

Predicting the range of substrates accepted by an enzyme from its amino acid sequence is challenging. Although sequence- and structure-based annotation approaches are often accurate for predicting broad categories of substrate specificity, they generally cannot predict which specific molecules will be accepted as substrates for a given enzyme, particularly within a class of closely related molecules. Combining targeted experimental activity data with structural modeling, ligand docking, and physicochemical properties of proteins and ligands with various machine learning models provides complementary information that can lead to accurate predictions of substrate scope for related enzymes. Here we describe such an approach that can predict the substrate scope of bacterial nitrilases, which catalyze the hydrolysis of nitrile compounds to the corresponding carboxylic acids and ammonia. Each of the four machine learning models (logistic regression, random forest, gradient-boosted decision trees, and support vector machines) performed similarly (average ROC = 0.9, average accuracy = similar to 82%) for predicting substrate scope for this dataset, although random forest offers some advantages. This approach is intended to be highly modular with respect to physicochemical property calculations and software used for structural modeling and docking.

On the intrinsic reactivity of highly potent trypanocidal cruzain inhibitors

The cysteine protease cruzipain is considered to be a validated target for therapeutic intervention in the treatment of Chagas disease. Then, we evaluated their potency against T. cruzi (Y and Tulahuen strains), which revealed the reversible cruzain inhibitor Neq0656 as a better trypanocidal agent (ECY50Y.strain = 0.1 mu M; SI = 58.4) than the current drug benznidazole (EC50Y.strain = 5.1 mu M; SI > 19.6). We also measured the half-life time by HPLC analysis of three lead compounds in the presence of glutathione and cysteine to experimentally assess their intrinsic reactivity. Results clearly illustrated the reactivity trend for the warheads (azanitrile > aldehyde > nitrile), where the aldehyde displayed an intermediate intrinsic reactivity. Clarification of the USP compendial procedure for phenoxybenzamine hydrochloride via updating impurity profiles

The current United States Pharmacopeia-National Formulary (USP-NF) and the British Pharmacopoeia phenoxybenzamine (PBA) hydrochloride drug substance and drug product monographs describe an HPLC procedure for the determination of a specified impurity tertiary amine phenoxybenzamine and use the resolution of an unknown related substance from PBA as a system suitability criterion; however, neither structural information of the unknown related substance is provided nor reference standards of the two impurities are available. PBA undergoes rapid degradation in neutral or basic aqueous solutions to generate the tertiary amine phenoxybenzamine as the predominant degradation product, which was confirmed as phenoxybenzamine hydroxide (PBA-OH). In addition, the unknown related substance was proposed as the phenoxybenzamine nitrile (PBA-CN) on the basis of LC-MS studies. The identity of PBA-CN was unambiguously verified via chemical synthesis, HPLC and NMR analyses. Studies on the pyrrolinone metabolites derived from the tobacco alkaloid 1-methyl-2-(3-pyridinyl)pyrrole (beta-nicotyrine)

Previous studies have established that the tobacco alkaloid 1-methyl-2-(3-pyridyl)pyrrole (beta-nicotyrine) is biotransformed by rabbit lung and liver microsomal preparations to an equilibrium mixture of the corresponding 3- and 4-pyrrolin-2-ones. Autoxidation of these pyrrolin-2-ones generates the chemically stable 5-hydroxy-5-(3-pyridinyl)-3-pyrrolin-2-one. This paper summarizes efforts to document more completely the pathway leading to this hydroxy-pyrrolinone. Chemical and spectroscopic evidence implicates the 2-hydroxy-1-methyl-5-(3-pyridinyl)pyrrole (2-hydroxy-beta-nicotyrine) as the key intermediate in this reaction pathway. Of potential toxicological interest is the detection of radical species derived from the autoxidation of this compound.
