Computational exploration of the 1,3-dipolar cycloaddition reaction of 7-isopropylidenebenzonorbornadiene with nitrile oxide and cyclic nitrone derivatives

The synthesis of isoxazolidine and isoxazole derivatives, versatile building blocks for the construction of a wide range of complex heterocyclic architectures in synthetic organic and medicinal chemistry, is efficiently achieved via the 1,3-dipolar cycloaddition reaction (1,3-DC). Herein, we report an extensive theoretical study on the peri-, regio-, stereo, and enantio-selectivities of 1,3-DC of 7-isopropylidenebenzonorbornadiene with nitrile oxide and cyclic nitrone derivatives using density functional theory calculations. Acetophenone-substituted nitrile oxide periselectively adds across the endocyclic olefinic bond of the dipolarophile to furnish the exo-cycloadduct as the major product, a reaction that has a rate constant of 1.88 x 10(9) s(-1). The endo approach of this periselective path is the closest competing pathway with a rate constant of 4.59 x 10(7) s(-1). Different substituents on the nitrile oxide do not affect the peri- and stereo-selectivity of the reaction. Diethyl ether solvation has no substantial effect on the energetic patterns observed in the gas phase computation. Also, we report a novel 1,3-DC between cyclic nitrone derivatives and 7-isopropylidenebenzonorbornadiene as an efficient way to generate isoxazolidine derivatives. Even though the reactions of the cyclic nitrone derivatives have slightly higher activation barriers than the acyclic nitrile oxide derivatives, the former is more enantioselective than the latter. Whereas electron-donating groups (EDGs) on the cyclic nitrone favor the formation of the exo-cycloadduct, electron-withdrawing groups (EWGs) favor the formation of the endo-cycloadduct. Both 1,3-dipoles add across the dipolarophile via a concerted asynchronous mechanism.

Simultaneous synthesis of thioesters and iron-sulfur clusters in water: two universal components of energy metabolism

Thioesters are important intermediates in both synthetic organic and biosynthetic reaction pathways. Here we show that thioesters can be synthesized in an aqueous reaction between thioacetate and thiols. The reaction can be coupled to a second reaction between sulfide and either ferrous or ferric iron, which drives the reaction forward. We furthermore demonstrate that sulfide released during thioester formation can be used in the synthesis of peptide bound [Fe-S] clusters, which like thioesters, are ancient components of metabolism. Together our results reveal a primordial linkage between high-energy ester formation and redox chemistry.

Sequential activation of a-bonds : Intermolecular cascade annulation with migration and remote functionalization

We demonstrate a new catalytic property of the lanthanide compound CeCl3 center dot 7H(2)O which sequentially activates N-H, sp(2) C-H and sp(3) C-H sigma-bonds at ambient temperature. The powerful catalyst enables selective C-C and C-N bond-forming intermolecular cascade annulation between enamines and diary1-1,2-diketone to afford functionalized 3,3-diaryl-3-pyrrolin-2-ones involving migration of an aromatic ring. We have also initiated a new multicomponent strategy utilizing readily available and inexpensive precursors and environmentally benign CeCl3 center dot 7H(2)O as an efficient catalyst under benign reaction conditions towards direct synthesis of several N-heterocycles including pentasubstituted 2-pyrrolinones and pyrroles. These observations provide new prospects and perspectives in sigma-bond activation process for easy access to functional molecules.

Amphiphilic dipyrrinones: methoxylated [6]-semirubins

Polymerization mechanism of 4-APN and a new catalyst for phthalonitrile resin polymerization

The widely used catalysts for phthalonitrile (PN) resin polymerization are aromatic compounds containing -NH2 because of their high catalytic performances. However, the catalytic mechanisms of these catalysts are not very clear. To understand the mechanisms of them, the widely used autocatalytic catalyst 4-(4-aminophenoxy)-phthalonitrile (4-APN) was studied in this paper. The polymerization process of 4-APN was tracked by a multi-purpose method, and ammonia gas was detected during the cross-linking processing for the fist time. Combined with the online IR results of the curing process of 4-APN, the mechanism of ammonia generation was newly proposed. Based on this mechanism, a new catalyst selection strategy was promoted, which is different from the traditional approach to catalyst selection for PN resin polymerization. According to the new strategy, 1,3-diiminoisoindoline (1,3-DII) was selected as a novel catalyst. The results showed that the new catalyst could not only effectively catalyze the polymerization of PN resin, but also has a lower curing temperature than that of organic amine catalysts and can eliminate the release of ammonia gas and the voids in the products caused thereby. Therefore, the results of this study will give important enlightenment to the development of PN catalysts and the development of PN.

Iodonium Cation-Pool Electrolysis for the Three-Component Synthesis of 1,3-Oxazoles

The synthesis of 1,3-oxazoles from symmetrical and unsymmetrical alkynes was realized by an iodonium cation-pool electrolysis of I-2 in acetonitrile with a well-defined water content. Mechanistic investigations suggest that the alkyne reacts with the acetonitrile-stabilized I+ ions, followed by a Ritter-type reaction of the solvent to a nitrilium ion, which is then attacked by water. The ring closure to the 1,3-oxazoles released molecular iodine, which was visible by the naked eye. Also, some unsymmetrical internal alkynes were tested and a regioselective formation of a single isomer was determined by two-dimensional NMR experiments.
