On the contrary, the yield of C5′-oxidation was found to be suppressed almost completely, while in homogeneous solutions it constituted approximately 14% of the total. The observed change in end product distribution is attributed to free valence transfer to and from the complexing agent, although the mechanisms associated with this process remain unclear. Transfected adenosine A(1) receptor-mediated modulation of thrombin-stimulated phospholipase C and phospholipase A(2) activity in CHO cells

Thrombin receptor activation in Chinese hamster ovary (CHO) cells stimulates the hydrolysis of inositol phospholipids and the release of arachidonic acid. Our previous studies have shown that activation of the human transfected adenosine A(1) receptor in CHO cells (CHO-A1) potentiates the accumulation of inositol phosphates elicited by endogenous P-2U purinoceptors and CCKA receptors. In this study we have investigated whether adenosine A(1) receptor activation can modulate thrombin-stimulated arachidonic acid release and/or inositol phospholipid hydrolysis in CHO-A1 cells. Thrombin stimulated [H-3]arachidonic acid release and total [H-3]inositol phosphate accumulation in CHO-A1 cells. Both these responses to thrombin were insensitive to pertussis toxin. The protein kinase C activator, phorphbol 12-myristate 13-acetate (PMA), potentiated thrombin-stimulated [H-3]arachidonic acid. In marked contrast, PMA inhibited thrombin-stimulated [H-3]inositol phosphate accumulation. The selective protein kinase C inhibitor Ro 31-8220 (3-{1-[3-(2-isothioureido)propyl]indol-3-yl}-4-(1-methylindol-3-yl)-3-pyrrolin-2,5-dione) had no effect on thrombin-stimulated [H-3]arachidonic acid release but reversed the potentiation of thrombin-stimulated [H-3]arachidonic acid release elicited by PMA. The selective adenosine A(1) receptor agonist N-6-cyclopentyladenosine (CPA) augmented the release of [H-3]arachidonic acid produced by thrombin. Co-activation of the adenosine A(1) receptor also potentiated thrombin-stimulated [H-3]inositol phosphate accumulation. The synergistic interactions between the adenosine A(1) receptor and thrombin were abolished in pertussis-toxin-treated cells. The potentiation of [H-3]arachidonic acid release by CPA was blocked by the protein kinase C inhibitors Ro 31-8220 and GF 109203X (3-[1-[3-(dimethylamino)propyl]-1H-indol-3-yl]-4-(1H-indol-3-yl)-1H-pyrrole-2,5-dione). In conclusion, thrombin receptor activation in CHO-A1 cells stimulates the accumulation of [H-3]inositol phosphates and the release of [H-3]arachidonic acid through pertussis-toxin-insensitive G-proteins. Experiments using PMA suggest that protein kinase C differentially regulates thrombin receptor activation of phospholipase C and phospholipase A(2). Co-activation of the transfected human adenosine A(1) receptor augments thrombin-stimulated phospholipase C and phospholipase A(2) activity. Temperature Dependence of Hyperfine Interaction for N-15 Nitroxide in a Glassy Matrix at 10-210 K

Principal N-15 hyperfine interaction (hfi) values in N-15-substituted nitroxide spin probe 3-carbamoyl-2,2,5,5-tetramethyl-3-pyrrolin-1-oxyl dissolved in nematic liquid crystal 4-pentyl-4′-cyanobiphenyl (5CB) were measured in a wide temperature range of 10-210 K, for 5CB frozen to a glassy state. X-band continuous-wave electron paramagnetic resonance (CW EPR) and pulse X- and Q-band N-15 electron-nuclear double resonance (ENDOR) techniques were employed. To avoid microwave saturation at low temperatures in CW EPR studies, a holmium complex Ho(Dbm)(3)Bpy (where Dbm is dibenzoylmethane and Bpy is 2,2′-bipyridine) was added. X-and Q-band N-15-ENDOR data have shown that the nitroxide hfi tensor is axially symmetric. The combination of data from all techniques allowed us to obtain the temperature dependence of isotropic and anisotropic parts of the nitroxide hfi tensor. Above similar to 100 K, a linear dependence of the anisotropic hfi value was observed, whereas below 30 K it was found to be nearly temperature independent. Such a behavior can be interpreted using the model of restricted orientational motions (librations) of a spin probe in a glassy matrix, with quantum effects occurring at low temperature (freezing of the librations). The energy quantum for the libration motion estimated from the temperature dependence of hfi of the spin probe is 84 cm(-1). Low-frequency Raman spectra of 5CB were also obtained, which provided the mean vibrational frequency of 76 cm(-1) for glassy 5CB.
