Synthetic Studies on Cyclocitrinol: Construction of the ABC Ring System Based on Epoxy-Nitrile Cyclization

The stereoselective synthesis of a model compound containing the ABC ring system of cyclocitrinol was accomplished. After connecting a C ring allyltitanium segment with an A ring bi-cyclo[4.1.0]heptanone segment, the seven-membered B ring moiety was constructed by an intramolecular cyclization reaction of an epoxy nitrile. The enone moiety was introduced through an oxidative decyanation reaction, and the bicyclo[4.4.1]undecane skeleton with the highly strained olefin moiety was formed through a ring-opening reaction of the bicyclo[4.1.0]heptane substructure.

Sprouting process enhances plant bioactive compounds. Broccoli (Brassica oleracea L) sprouts are well known for their high levels of glucosinolates (GLs), amino acids, and antioxidants, which offer outstanding biological activities with positive impacts on plant metabolism. Elevated CO2 (eCO(2), 620 ppm) was applied for 9 days to further improve nutritive and health-promoting values of three cultivars of broccoli sprouts i.e., Southern star, Prominence and Monotop. eCO(2) improved sprouts growth and induced GLs accumulation e.g., glucoraphanin, possibly through amino acids production e.g., high methionine and tryptophan. There were increases in myrosinase activity, which stimulated GLs hydrolysis to yield health-promoting sulforaphane. Interestingly, low levels of ineffective sulforaphane nitrile were detected and positively correlated with reduced epithiospecifier protein after eCO(2) treatment. High glucoraphanin and sulforaphane levels in eCO(2) treated sprouts improved the anticarcinogenic and anti-inflammatory properties of their extracts. In conclusion, eCO(2) treatment enriches broccoli sprouts with health-promoting metabolites and bioactivities.

The (spectro)electrochemical behavior of 4a,b and 6 was studied at 25 and -72 degrees C and the results thereof are compared with [NEt4][RuCl4(N equivalent to CMe)(2)] (7b) and [RuCl(N equivalent to CPh)(5)][PF6] (8). The electrochemical response of the cation and the anion in 4a,b are independent from each other. [RuCl(N equivalent to CR)(5)](+) possesses one reversible Ru-II/Ru-III process. However, [RuCl4(N equivalent to CMe)(2)](-) was shown to be prone to ligand exchange and disproportionation upon formation of either a Ru-IV and Ru-II species at 25 degrees C, while at -72 degrees C the rapid conversion of the electrochemically formed species is hindered. In situ IR and UV/Vis/NIR studies confirmed the respective disproportionation reaction products of the aforementioned oxidation and reduction, respectively.

The toxicity of germander, a herb used to treat obesity, is attributed to cytochrome P450 activation of the furan ring of its major diterpenoid component (teucrin A) into a reactive metabolite capable of adducting proteins. 1,4-Enedials have been proposed to be the reactive products of metabolism, possibly arising from a rearrangement of putative epoxide intermediates. We synthesized the enedial derivative of teucrin A as well as the enedial derived from a model furan, 3-(4-methoxy-benzyloxymethyl)-furan, by dimethyldioxirane oxidation and characterized the products of their reactions with amino acids and peptides. The reactions of the model enedial, 2-(4-methoxy-benzyloxymethyl)-but-2-enedial,withN-acetyl lysine (NAL) afforded regioisomeric N-alkyl-3-pyrrolin-2-ones, differing in the substitution on the double bond of the heterocyclic ring. Novel products formed in the reactions of the model enedial with N-acetyl cysteine (NAC) and both NAC/NAL uncovered the existence of tautomerization between the enedial and a hydroxyenal, which was manifest by the loss of 4-methoxybenzylalcohol and the incorporation of a second molecule of NAC. The reactions of teucrin A-enedial with NAC and NAL afforded analogues of the products observed with the model enedial, and the existence of the tautomeric equilibrium resulted in epimerization of the proton (H12) adjacent to the former furan ring. This work further illuminates the complex chemical behavior of unsaturated dialdehydes as an important class of toxic metabolites and lays the foundation for studies of the protein targets of teucrin A-enedial.

Energetically significant nitrile center dot center dot center dot nitrile and unconventional C-H center dot center dot center dot pi(nitrile) interactions in pyridine based Ni(II) and Zn(II) coordination compounds: Antiproliferative evaluation and theoretical studies

Two new coordination compounds viz. [Ni(2,6-PDC)(Hdmpz)(H2O)(2)]center dot H2O (1) and [Zn(3-CNpy)(2)Cl-2] (2) (2,6-PDC = 2,6-pyridinedicarboxylate, Hdmpz = 3,5-dimethylpyrazole, 3-CNpy = 3-cyanopyridine) have been synthesized and characterized using elemental analysis, thermogravimetric analysis, electronic, infrared spectroscopy and single crystal X-ray diffraction techniques. Crystal structure analyses reveal the presence of supramolecular assemblies involving interesting dimers with unconventional contacts in the compounds. DFT (Density Functional Theory) calculations on the supramolecular dimers in the crystal structure of 1 reveal that the sum of contributions of anion-pi, pi-pi and other long range interactions due to the approximation of the bulk monomers is energetically significant. Molecular Electrostatic Potential (MEP) surface and Quantum Theory of Atoms in Molecules (QTAIM) analyses on the interesting supramolecular dimers of the crystal structures of 2 reveal the presence of unconventional anion center dot center dot center dot pi contacts involving coordinated chlorido ligands and C-H center dot center dot center dot pi(nitrile) interactions involving the pi-system of the nitrile moiety of 3-cyanopyridine. Remarkably, Atoms in Molecules analysis also confirms the existence of energetically significant unconventional anti-parallel nitrile center dot center dot center dot nitrile interaction in the crystal structure of 2. Cell cytotoxicity of the compounds performed in Dalton’s lymphoma (DL) malignant cancer cell line showed effective potency with negligible cytotoxicity in normal cells (similar to 12%). It is interesting that compound 1has excellent cytotoxic potency with IC50 closer to cisplatin and can bind different biological targets with similar signalling pathways.
