Comparative study of fluxional processes at two different classes of eight-coordinate rhenium(V) polyhydride complexes

Two different structural types of dodecahedral rhenium(V) tetrahydride complexes were identified as capable of providing insight into a pseudorotational exchange of ligands at such complexes. One type of complex afforded an inequivalent set of NMR-active phosphorus atoms that occupy B sites in the rhenium coordination sphere along with four hydride ligands in the dodecahedral A sites. A second type of complex afforded the potential to inhibit the pseudorotational exchange of ligands at such complexes due to the presence of a chelating bidentate ligand. A pair of each type of complexes were studied by variable temperature NMR measurements and simulation of those measurements. The first type of complex exhibited pseudorotation of both the A site hydride ligands and the B site triphenylphosphine ligands. The first type of complexes also exhibited a pairwise exchange of the two inequivalent hydride ligands on the complexes. The second type of complex did not exhibit pseudorotational exchange of the A site hydride ligands, presumably due to the chelating bidentate ligand bound to rhenium. Instead, the A site hydride ligands of the second type of complex exhibited only exchange with the single B site hydride ligand through either a pairwise or a turnstile type of mechanism. Neither type of complex exhibited any direct intermolecular exchange of protons from the hydride ligand set with adventitious water from the solvent matrix. The value of Delta G(240)(double dagger) for each of the identified fluxional processes was close to 12 kcal/mol. The values of Delta S-double dagger for each fluxional process suggest that none of the processes are dissociative in nature.

A Nanomechanical Study on Deciphering the Stickiness of SARS-CoV-2 on Inanimate Surfaces

The SARS-CoV-2 virus that causes the COVID-19 epidemic can be transmitted via respiratory droplet-contaminated surfaces or fomites, which urgently requires a fundamental understanding of intermolecular interactions of the coronavirus with various surfaces. The corona-like component of the outer surface of the SARS-CoV-2 virion, named spike protein, is a key target for the adsorption and persistence of SARS-CoV-2 on various surfaces. However, a lack of knowledge in intermolecular interactions between spike protein and different substrate surfaces has resulted in ineffective preventive measures and inaccurate information. Herein, we quantified the surface interaction and adhesion energy of SARS-CoV-2 spike protein with a series of inanimate surfaces via atomic force microscopy under a simulated respiratory droplet environment. Among four target surfaces, polystyrene was found to exhibit the strongest adhesion, followed by stainless steel (SS), gold, and glass. The environmental factors (e.g., pH and temperature) played a role in mediating the spike protein binding. According to systematic quantification on a series of inanimate surfaces, the adhesion energy of spike protein was found to be (i) 0-1 mJ/m(2) for hydrophilic inorganics (e.g., silica and glass) due to the lack of hydrogen bonding, (ii) 2-9 mJ/m(2) for metals (e.g., alumina, SS, and copper) due to the variation of their binding capacity, and (iii) 6-11 mJ/m(2) for hydrophobic polymers (e.g., medical masks, safety glass, and nitrile gloves) due to stronger hydrophobic interactions. The quantitative analysis of the nanomechanics of spike proteins will enable a protein-surface model database for SARS-CoV-2 to help generate effective preventive strategies to tackle the epidemic.

ULTRASOUND IN FATTY-ACID CHEMISTRY – SYNTHESIS OF A 1-PYRROLINE FATTY-ACID ESTER ISOMER FROM METHYL RICINOLEATE

A novel 1-pyrroline fatty acid ester isomer [viz. 8-(5-hexyl-1-pyrrolin-2-yl)octanoate] has been synthesized from methyl ricinoleate by two routes with an overall yield of 42 and 30%, respectively. Most of the reactions are carried out under concomitant ultrasonic irradiation (20 KHz, ca. 53 watts/cm(2)). Under such a reaction condition, the reaction time is considerably shortened, and product yields are high. Dehydrobromination under concomitant ultrasonic irradiation of methyl 9,10-dibromo-12-hydroxyoctadecanoate with KOH in EtOH furnishes methyl 12-hydroxy-9-octadecynoate (66%) within 15 min. Hydration of the latter under ultrasound with mercury(II)acetate in aqueous tetrahydrofuran yields exclusively methyl 12-hydroxy-9-oxo-octadecanoate (95%) in 30 min. The hydroxy group in the latter compound is transformed to the azido function via the mesylate, and treatment of the azido-oxo intermediate (methyl 12-azido-9-oxooctadecanoate) with Ph(3)P under ultrasonic irradiation furnishes the requisite 1-pyrroline fatty acid ester (77%). The same azido-oxo intermediate has also been obtained by the oxidation of methyl 12-azido-9-cis-octadecenoate using benzoquinone and a catalytic amount of Pd(II)chloride in aqueous tetrahydrofuran under concomitant ultrasonic irradiation (90 min) to give the product in 45% yield. The latter reaction does not take place even the reaction mixture.

The electrochemical response of the cation and the anion in 4a,b are independent from each other. [RuCl(N equivalent to CR)(5)](+) possesses one reversible Ru-II/Ru-III process. However, [RuCl4(N equivalent to CMe)(2)](-) was shown to be prone to ligand exchange and disproportionation upon formation of either a Ru-IV and Ru-II species at 25 degrees C, while at -72 degrees C the rapid conversion of the electrochemically formed species is hindered. In situ IR and UV/Vis/NIR studies confirmed the respective disproportionation reaction products of the aforementioned oxidation and reduction, respectively.

Molecular modeling, synthesis, and structures of N-methylated 3,5-linked pyrrolin-4-ones toward the creation of a privileged nonpeptide scaffold

The molecular modeling, synthesis, and elucidations of the solid state and solution structures of N-methylated 3,5-linked bispyrrolin-4-ones are described. Prior investigations established that the 3,5-linked pyrrolin-4-one based scaffold can be incorporated into mimics of beta-sheet/beta-strands and into potent, orally bioavailable inhibitors of the HIV-1 protease. To extend the utility of this scaffold beyond that of the initially designed mimics of beta-sheet/beta-strands, we have now explored the structure of N-methylated pyrrolinones. Molecular modeling indicated that N-methylated bispyrrolinones could adopt three low-energy backbone conformations (ca. 165 degrees, 289 degrees, and 320 degrees). Upon their successful synthesis, structural elucidation both in the solid state and in solution revealed the existence of two of the three predicted backbone conformers (ca. 165 degrees and 289 degrees). Two structures were particularly noteworthy and completely unexpected. Mono-N-methyl bispyrrolinone (+)-1 self assembled in the solid state to form a novel helix, while the acetylene-linked dimer of(+)-1, designed to potentiate the observed helical array, instead associated via an intermolecular hydrogen bond in parallel columns. These serendipitous observations led us to speculate that the pyrrolinone moiety may in fact represent a privileged nonpeptide scaffold, able to mimic not only the extended beta-sheet/beta-strand conformation as initially targeted, but also diverse conformations including those analogous to beta-turns and helices. These seemingly unlimited conformations greatly expand the scope of this scaffold for the development of low-molecular weight ligands for biologically important macromolecules.
