Simultaneous synthesis of thioesters and iron-sulfur clusters in water: two universal components of energy metabolism

Thioesters are important intermediates in both synthetic organic and biosynthetic reaction pathways. Here we show that thioesters can be synthesized in an aqueous reaction between thioacetate and thiols. The reaction can be coupled to a second reaction between sulfide and either ferrous or ferric iron, which drives the reaction forward. We furthermore demonstrate that sulfide released during thioester formation can be used in the synthesis of peptide bound [Fe-S] clusters, which like thioesters, are ancient components of metabolism. Together our results reveal a primordial linkage between high-energy ester formation and redox chemistry.

Sequential activation of a-bonds : Intermolecular cascade annulation with migration and remote functionalization

We demonstrate a new catalytic property of the lanthanide compound CeCl3 center dot 7H(2)O which sequentially activates N-H, sp(2) C-H and sp(3) C-H sigma-bonds at ambient temperature. The powerful catalyst enables selective C-C and C-N bond-forming intermolecular cascade annulation between enamines and diary1-1,2-diketone to afford functionalized 3,3-diaryl-3-pyrrolin-2-ones involving migration of an aromatic ring. We have also initiated a new multicomponent strategy utilizing readily available and inexpensive precursors and environmentally benign CeCl3 center dot 7H(2)O as an efficient catalyst under benign reaction conditions towards direct synthesis of several N-heterocycles including pentasubstituted 2-pyrrolinones and pyrroles. These observations provide new prospects and perspectives in sigma-bond activation process for easy access to functional molecules.

The efficiency of Overhauser dynamic nuclear polarization (DNP) depends on the local dynamics modulating the dipolar coupling between the two interacting spins. By attaching nitroxide based spin labels to molecules and by measuring the H-1 DNP response of solvent water, information about the local hydration dynamics near the spin label can be obtained. However, there are two commonly used types of nitroxide ring structures; a pyrroline based and a piperidine based molecule. It is important to know when comparing different experiments, whether changes in DNP enhancements are due to changes in local hydration dynamics or because of the different spin label structures. In this study we investigate the key parameters affecting DNP signal enhancements for 3-carbamoyl-2,2,5,5-tetramethyl-3-pyrrolin-1-oxyl, a 5-membered ring nitroxide radical, and for 4-oxo-2,2,6,6-tetramethyl-1-piperidinyloxy, a 6-membered ring nitroxide radical. Using X-Band DNP, field cycling relaxometry, and molecular dynamics simulations, we conclude that the key parameters affecting the DNP amplitude of the H-1 signal of water to be equal when using either nitroxide. Thus, experiments measuring hydration dynamics using either type of spin labels may be compared. (C) 2009 Elsevier Inc. All rights reserved.

A one-step synthesis of 5-alkoxypyrrole-3-phosphonates is presented starting from suitable alpha-cyanomethyl-beta-ketophosphonates. The key step in the synthesis involves a one-pot addition and heteroannulation sequence. The zinc perchlorate-catalyzed addition of alcohols to the nitrile carbon of alpha-cyanomethyl- beta-ketophosphonates followed by annulation furnished 5-alkoxypyrrole-3-phosphonates. Replacing the typical beta-alkyl substituents of [6]-semirubin and [6]-oxosemirubin, two intramolecularly hydrogen-bonded bilirubin analogs, with methoxy groups produces amphiphilic dipyrrinones. In contrast, in the solid, X-ray crystallography reveals supramolecular ribbons of intermolecularly hydrogen-bonded (dipyrrinone to dipyrrinone and acid to acid) 2,3,7,8-tetramethoxy-[6]-semirubin.
